Application Notes

HEPES buffer commonly used in cell culture media. Theaddition of 10–25 mM HEPES provides extra bufferingcapacity when cell culture requires extended periods ofmanipulation outside of a CO<sub>2</sub> incubator. HEPESis a good buffering choice for many cell culture systemsbecause it is membrane impermeable, has limited effect onbiochemical reactions, is chemically and enzymatically stable,and has very low visible and UV light absorbance. HEPES isthe buffer of choice in a protein deposition technique inelectron microscopy because it did not affect metal substrates.HEPES has been evaluated and shown to be suitable for usewith Ampholines in generating pH gradients less than 1 pHunit wide for isolectric focusing applications. HEPES is therecommended buffer for the glutamate binding assay becauseit prevents binding to non-receptor materials.

Preparation Method

A buffer solution of HEPES can be prepared by any of severalmethods. The free acid can be added to water, then titratedwith approximately one-half mole equivalent of sodiumhydroxide or potassium hydroxide to the desired pH. A simplemixing table for preparing 0.05 M HEPES/NaOH has beenpublished. Alternatively, equimolar concentrations of HEPESand of HEPES sodium salt can be mixed in approximatelyequal volumes and back-titrate with either solution to theappropriate pH. The sodium salt can be titrated with HCl toyield a half-equivalent of sodium chloride; however, theaddition of the ionic strength will change the osmolality of thesolution. Most solutions of HEPES hemisodium will dissolve inwater forming a pH at 7.5 with little to no adjustment.
