Neuroinflammation

•  Neuroinflammationisinflammationofthenervoustissuewhichmaybeinitiatedinresponsetoavarietyortriggersincludinginfection,traumaticbraininjury,toxicmetabolitesorautoimmunity

•  IntheCNSwhichincludesthebrainandthespinalcord,microgliaaretheresidentinnateimmunecellsthatareactivatedinresponsetothesetriggers

•  TheCNSistypicallyanimmunologicallyprivilegedsitebecausetheperipheralimmunecellsaregenerallyblockedbythebloodbrainbarrier,aspecializedstructurecomposedofastrocytesandendothelialcells.

•  CirculatingperipheralimmunecellsmaycrossacompromisedBBBandencounterneuronsandglialcellsexpressinghistocompatibilitycomplexmoleculeswhichperpetuateanimmuneresponse

•  ThisresponseisdesignedtoprotecttheCNSfromtheinfectiousoffendingagentsbutmaybetoxicandcreatewidespreadinflammationaswellasfurthermigrationofleukocytesthroughtheBBB

• NeuroinflammationwasonceaclearlydefinedtermdenotingpathologicalimmuneprocesseswithintheCNS.

• Thefourhallmarksofperipheralinflammation:CNSinjuries,stroke,injuryorinfection

• Recentlythedefinitionofneuroinflammationhasrelaxedtothepointthatitisoftennowassumedtobepresentwhenevenonlyasingleclassicalhallmarkofinflammationwasmeasured.

• Unexpectednon-classicalimmuneactionsofimmunemediatorsandcellsintheCNSintheabsenceofpathologyhaveincreasedthelikelihoodthathomeostaticandadaptiveimmuneprocessesintheCNSwillbemistakenforneuroinflammation.
