The ELITech Process for Gram StainingThe Aerospray stainer stainsmicroscope slides smeared withmaterials containing micro-organ-isms such as culture materials,body fluids, or exudates. ELITechgram reagents are tailored to givesuperb results in the Aerospraystainer.
HOW IT WORKSReagents are applied from atom-izing spray nozzles onto micro-scope slides containing the speci-mens. Staining is rapid and precise. Specimens contact onlyfresh stain, avoiding specimencross-contamination or contactwith contaminated reagents.
FIXATIONSpecimens can be heat-fixed andstained directly, or the automaticfixation feature can be used toprovide convenient and reproducible specimen fixation.
PRIMARY STAINOnce slides are fixed, they aresprayed with crystal violet, a pos-itively charged (basic) dye whichrapidly penetrates microbial cells
and stains them dark blue-to-black. The excess stain is thenspun off and washed away withdeionized water to prevent pre-cipitate formation outside stainedcells.
IODINEIodine reagent is then sprayedonto the slide. The iodine pene-trates the cells and forms a precipitate (mordant) with thecrystal violet dye. Excess iodineis spun off and rinsed from theslide with deionized water.
DECOLORIZING ANDCOUNTERSTAINDecolorizer is then applied toremove the crystal violet-iodinecomplex until only the gram-neg-ative organisms are decolorized.Safranin or fuchsin (contained inthe decolorizer) simultaneouslycounter-stains the gram-negativecells pink-to-red.
Decolorization is the critical stepin proper gram differentiationand unless properly controlledcan lead to errors. The Aerospray
stainer provides a choice ofdecolorizer cycles ranging from 1to 9, allowing you to compensatefor thick specimens, or other pos-sible variables. With the ELITechsystem, you can reproduciblydifferentiate sensitive organismsin normal smears.
With its rapid staining cycle andhigh throughput capability, theAerospray stainer is a convenient,cost effective way to increasegram staining capacity.
DRYINGThe slides are spun at about 950RPM for 45 seconds. This drawsair through the stainer and driesthe slides. Slides emerge from thestainer dry and ready for themicroscope.
With its rapid staining cycle andhigh throughput capability, theAerospray stainer is a convenient,cost effective way to stain gramspecimens.
