The emphasis of my research is the mechanisms regulating gene expression during fruit ripening and development. Additionally I am interested in discovery of genes that control desirable attributes of fruits. The projects being perused in my laboratory include identifying genes that regulate fruit shelf-life, texture, ripening and processing quality. Other important focus of his lab is to identify mechanisms that regulate levels of antioxidants, minerals and vitamins in ripening fruits. We are also perusing role of molecular modification in enhancing antigenicity of vaccine molecules. Some of recent accomplishments include: 1) Engineering over 3-fold increase in the levels of lycopene in tomato fruits. Lycopene has been the focus of much attention since 1995, as it has been shown to reduce risk of developing prostate cancer by 45 % and several other cancers by about 20 %. 2) Modifying pectin chemistry by down-regulating levels of pectin methylesterase and polygalacturonase in tomato to enhance juice viscosity and

net processing yield. 3) Characterization of heat stress response in order to improve heat tolerance and pollination in tomato. We have identified and characterized effects of a novel small heat shock protein gene, designated as vis1 (viscosity 1), that regulated tomato juice viscosity. 4) Mechanisms regulating homeostasis in fruit to maintain fruit texture. Our results show that down regulation of a fruit ripening associated lipoxygenase reduces ion leakage and enhances fruit firmness (US patent 6355862). 5) Expressed avian flu vaccine gene in both tomato and tobacco plants and cell cultures. We have developed a tomato fruit macroarrays to characterize global changes in fruit gene expression under various growth conditions, including stress. We are using this array to characterize gene whose expression is modified in ‘yellow shoulder disorder in tomato. Other interests include developing and characterizing a transposon insertion mutant population of Microtom, a dwarf tomato variety. This project would likely identify many genes of potential interest to improve production and overall quality, including levels of vitamins and mineral, in fruit and vegetable crops.
