The EXCEL spreadsheet shows an example of collected data obtai-ned with the bbe Cuvette Fluorometer. Water samples were takenfrom water surface at the same place off the shore of Lake Bistenseeduring July 1999. Data was stored in the internal data logger ofbbe Cuvette Fluorometer. Increasing sunlight with rising temperatu-res lead to enhanced growth of blue-green algae while concentra-tion of other groups was not largely affected. Developing bloomscan therefore be detected early on.

The bbe Cuvette Fluorometer is a well established instrument for chlorophyllanalysis. For single measurements or measurement of collected water sam-ples the fluorometer provides prompt and reliable results of algae content.Based on fluorescence excitation spectra up to 5 different algae classes aredetermined simultaneously. No preceding treatment is needed. Measureddata is stored in the integral data logger or sent to a connected PC.
Excitation of algae by visible light results in fluorescence from chlorophylla, a pigment found in all photosynthetic reaction centres of algae andother photosynthetic organisms. Incidence of other pigments like phycocya-nin or fucoxanthine are typical for different algae classes. Interaction ofthese different pigment systems with a chlorophyll a core results in specificexcitation spectrum for a taxonomic related algae class. The specific pat-terns of algae fluorescence – the fingerprints – are used with the bbe soft-ware for quantification of the different classes. A set of 5 LED with selectedwavelengths are used for sequenced illumination. Fingerprints of newly intro-duced algae can be recorded for a more detailed analysis.
Optional information about photosynthetic activity can be achieved.Referring to the Genty-Parameter fo, f, fm fluorescence measurement in aprogrammed mode enables calculation of active chlorophyll under definedconditions. This measurement is comparable to measurement of oxygenevolution by photosynthetic active membranes. An optional toxicity test soft-ware is available for detection of herbicides.
Easy operation
The approved software developed for bbe Cuvette Fluorometer is used tocontrol the algae analyzer. Quantitative distribution of algae class are perma-nently processed and displayed. Collected data can be easily transferred toconnected PC via RS232 serial communications. Start, stop and operation ofthe Fluorometer is achieved via a simple membrane keypad on top of instru-ment. A selfexplanatory menu enables rapid access to all important functionsand operations.
