The enzyme-linked immunospot(ELISPOT) assay is a powerful tool fordetecting and enumerating individualcells that secrete a particular protein in vitro. Based on the sandwichenzyme-linked immunosorbent assay(ELISA), the ELISPOT assay derives itsspecificity and sensitivity by employinghigh affinity capture and detectionantibodies and enzyme-amplification.Originally developed for analyzing specific antibody-secreting cells, theassay has been adapted for measuringfrequencies of cells that produce andsecrete other effector molecules, suchas cytokines. The sensitivity of theassay lends itself to measurement ofeven very low frequencies of cytokine-producing cells (e.g., 1/300,000).Recent developments in assay platedesign and in ELISPOT plate-readerinstrumentation have significantlyimproved the utility of the ELISPOTmethod for objective and rapid analysisof cytokine-producing cells.

BD Biosciences Pharmingen is pleased to announce the availability of optimized ELISPOT reagent sets and instrumentation, in collaborationwith Cellular Technology Ltd (CTL) of Cleveland, OH. BD BiosciencesPharmingen is now marketing CTL’sstate-of-the-art automated ELISPOTplate reader, the ImmunoSpot™ Series 1Analyzer, along with high sensitivityImmunoSpot™ ELISPOT plates andELISPOT-optimized antibody pairs. The ImmunoSpot™ Analyzer offers a degree of sensitivity and specificitywhich is not possible using otherapproaches, as well as unique featuresfor data analysis, storage and highthroughput screening.

Primed human PBMCs were restimulated in culture

with PMA and ionomycin in the microwell of an

ImmunoSpot™ ELISPOT plate, which was precoated

with unlabelled NA/LE™ anti-human IL-10 antibody

(Human IL-10 ELISPOT Reagent Set, Cat. No. 551018).

Biotinylated anti-human IL-10 antibody was used to

detect the captured IL-10. Spots were visualized using

avidin-HRP enzyme and AEC substrate, followed by

image analysis and spot enumeration using an

ImmunoSpot.™ The spot size distribution of the PMA

and ionomycin-induced response, as measured

by image analysis, is shown in Panel B.

Developed for the highest accuracy with serum, plasma, and other biologicalfluids, the BD OptEIA™ ELISA Kit II format is the latest product of BD Biosciences Pharmingen’s ongoing assay R&D efforts. The Kit II formatdemonstrates mean spike recovery improvements of 7% to 41% in serum and plasma. With cell culture supernatant samples, values in the original kitsand the Kit II’s are equivalent. The BD OptEIA™ Kit II contains a new ELISA diluent, standard/sample diluent, and one-step TMB substrate reagent,with an improved protocol.

The antibodies used in the BD OptEIA™ Kits are specifically fragmented to enable accurate quantitation of cytokines and other soluble proteins in serum, plasma, and cell culture supernatant samples. Two pre-coated plates and all necessary reagents are provided.
