Bio-Luminescence Detection
Project Manager: Eitan NovotnySean Dang

Overview
• Biological rhythms– Samples emit periodic light– Periods around 24 hours– Sample rate once every 10 minutes
• Commercial device flaws– Missing flow through function– Product is costly
• Design goals– Integration with original equipment– Improve function and reduce cost
• MDR Specification• Design Changes
Box Design
• 39cmx39cmx32cm• Designed in Pro
Engineering• Two Pieces
– Main box– Lid (top and front)
Box Design - Details
• Internal temperature must match incubator
• Two 80mm square holes plus one 80mm fan for airflow
• Light isolated environment
• 5 BNC connectors for PMTs and Motor
• 2 18cmx4cm windows for tube baffles
Box Design - Overview
• Light baffle
Main Box
Lid
Motor
• 1240i board works with specific motors
• Parallel motor connection not possible
• Principal needs– Single high torque motor– Four rotating mirrors
Motor - Solution
• Motor is Oriental Motor PK243A1A-SG18– Used in commercial application
• Step up gear used for high precision
• Central gear and outer gear are 5”
• Shafts connect outer gears to mirrors
• Contact switches used for error checking
Light Source/Detector
• Luciferin - emits light• Wavelength range: 268nm
to 537nm• Photomultiplier Tube
(PMT)• Detects wavelength range
from 185nm to 680nm• Effective area 4mm x
20mm
Light Efficiency/Path Length
• Commercial application has a .25 cm path length
• Our light path length is about 3cm to 5cm
• Light goes to the mirror and reflects up to the PMT
• Loss of light due to vertical arrangement
• To minimize loss of light, add mirrors
Light Efficiency - Continued
• Interference from other light sources
• Add reflective walls, to minimize interference from other light sources
Electronics
• H8259 from Hamamatsu
• PCI-6601 Counter Board
• Applied Motion 1240i
• Oriental Stepper Motor
• Computer and software
BudgetPart Cost (each) Qty Price
• H8259 $1150 4 $4600• 1240i $200 1 $200• PCI-6601 $600 1 $600• Motor $177 1 $177• Metal $46.11 6 $277• Gears $62.02 5 $310• Shaft Holder $27.46 5 $137• Shaft $20.05 1 $20• Wires $100 $100
Total* $6421*total cost to date
Conclusion
• MDR Specification done before 2nd semester
• Accomplishments– Gearing design done– Box design done
• Mirror design will be completed before conclusion of semester
• Device should be completed on time
• Device should be under budgetAluminum for box construction
