Fluid dynamical results (1)
In the control group (n=5) with its fully transformed type I fibered muscle after 6 months of chronic electrical stimulation, stroke volumina between 10 to 15 ml could be measured. With a maximum contraction frequency of 30 beats per minute, a skeletal muscle ventricle output of 300 to 450 ml per minute is achieved in a permanent manner (mean stimulation frequency: 30bpm x 6 Pulse/Burst
3 Hz)
In group II (n=5) with 6 months of intermittened stimulation (day: 15 min on, 15 min off, night: off) and a mean stimulation frequency ~ 0,7 Hz, stroke volumina between 35 und 50 ml could be measured. At a frequency of 30 beats per minute, an output of up to 1,5 Liters was reached. Higher loading of the skeletal muscle ventricle was possible, but after 2 to 5 minutes, a reversible decline of the output to 50% and less was observed.

Ca2+-Freisetzung aus dem sarkoplasmatischen

a). Die Interaktion von S100A1 mit Myofilamenten
reduziert deren Rigidität b), was zu einer gesteigerten Compliance
des Ventrikels
führt. Die Calciumsensitivität der Myofilamente
wird durch S100A1 herabgesetzt c). Dieses erlaubt eine schnellere diastolische Dissoziation des Calciums, mit der Folge einer gesteigerten
Lusitropie. S100A1 hebt die inhibitorische
Wirkung von Phospholamban
(PLB) auf die Sarkoendoplasmatische-
Retikulum-Calcium-ATPase
(SERCA) auf d), daraus resultiert eine verbesserte Calciumresequestration. Des Weiteren minimiert S100A1 die Öffnungswahrscheinlichkeit des RyR2 in der Diastole und verringert so das sarkoplasmatische
„Calciumleck“
e). In den Mitochondrien der Herzmuskelzelle wird durch S100A1 die Produktion von ATP gesteigert f).
