Dear Riders, Volunteers and Sponsors,

The 2014 PMC goes down in our 35-year history as the coldest and wettest ever. While the conditions undoubtedly tasked individual will, for me the lasting memory from PMC 2014 is the spirit and pride of our committed PMC community. I love a cool, 75-degree, sunny riding day as much as the next, but I have always learned much more from my endeavors in the rain. While the mission to raise funds for cancer research is at the center of our work, the PMC community is a shining example of the best of human nature. A project of this scope could not be achieved without more than 3,500 volunteers who covered 360 miles of infra-structure. Yet, their jobs were made immeasurably easier because of the character of the people on bicycles, whose good nature and humor warmed our hearts even as our fingers and toes froze. Despite the weather, at $41 million raised, we surpassed our own fundraising goal and increased our 35-year Jimmy Fund total contribution to $455 million. Thank you all. As you reminisce over the following pages, please note the many smiles captured through the rain. I suspect the love, camaraderie, and resolve to overcome brought life lessons we will refer to for years to come. Thank you for your continued commitment to the PMC.

The Pan-Mass Challenge remains a significant force behind Dana-Farber Cancer Institute’s fight against cancer, raising $455 million since 1980 to bolster priorities at the Institute that are most in need. Empowered by the PMC’s unrestricted flexible funds, Dana-Farber continues to make great progress in the delivery of precision medicine for patients everywhere. An Inside the Park Home Run!The PMC Raises $41 Million for Dana-Farber Cancer Institute

From a magically lit home plate at Fenway Park on the evening of November 8th, Billy Starr addressed hundreds of PMC riders and guests gathered in the stands. Umbrella held overhead in tribute to the 35th year’s rainy ride, Starr directed guests’ attention to the park’s jumbotron where a record gift of $41 million to Dana-Farber Cancer Institute was unveiled. The 35th ride was an amazing success. This incredible total was raised by 5,498 cyclists who participated in one of the PMC’s twelve routes on August 2nd and 3rd. The annual bike-a-thon attracts riders from across the world, over 300 of whom are cancer survivors or cur-rent patients. Cyclists commit to raising between $500 and $5,000, minimums exceeded by ninety percent of riders; one-third raise more than twice the amount required. “Our riders are incredibly passionate about riding and finding cures for cancer,” said Starr. “We all share a special camaraderie, built by the shared experiences of training, riding and understanding the impacts of cancer person-ally or otherwise. It’s this bond that makes our unprecedented achievements in support of can-cer research and Dana-Farber so extraordinary.” With 100% of every rider-raised dollar going directly to the Jimmy fund, 2014’s $41 million gift brings the PMC’s total donated to the institute to $455 million since 1980.

Rain? That’s Just Puddle-Stomping for Living Proof Cyclists and Volunteers Of all the PMCers, it might have been easiest for the 584 cancer survivors, known as PMC Living Proof cyclists and volunteers, to say “pshaw” to the driving rain and cold on Saturday of PMC weekend. Many Living Proofers have beat cancer; some ride while on treatment regimens; others recently finished treat-ment. On PMC Saturday, they rode up to 110 miles or volunteered in the cold rain. Then they gathered at the Mass Maritime Academy in Bourne overlooking the bay and the Sagamore Bridge for the celebratory Living Proof photo. With WBZ-TV’s radar truck tracking the another imminent squall, PMC riders and volunteers toasted their health and celebrated another year with PMC.

The Boston Red Sox Foundation, Official Charity of the Boston Red Sox, has been a PMC presenting sponsor since 2003. In that time the Red Sox have won three World Series trophies. An incredible run, no less credible than the $327,584,000 the PMC raised for DFCI during that time. And the founda-tion’s support for the PMC event oper-ations is one big reason the PMC’s gift represents 100% of what every rider raised. The Red Sox support extends beyond sponsorship. The PMC logo is displayed on the Green Monster and they field a PMC team that has raised almost $3.5 million. Stacey Lucchino, wife of Red Sox President and CEO Larry Lucchino, captains team Nine. Larry said, “The PMC is a cause near and dear to each of us, and the part-nership that the Red Sox are lucky to share with this bike-athon is unlike any other in sports.”

One Lung and Training Wheels Got Living Proof Cyclist Michelle Sanders in the Saddle As an infant, Michelle Sanders was diag-nosed with neuroblastoma, a cancerous tumor that started in her spinal cord and wrapped around her right lung. The year was 1969, when neuroblastoma at any age was consid-ered a death sentence. She underwent surgery and radiation, which permanently damaged her right lung. Over the years, Sanders had numerous other surgeries to manage the after-effects of her cancer treatment.

To help Sanders cope, her parents gave her a bicycle. That was just before third grade, and she picked out a blue Raleigh Mountie three-speed. She flew around the neighborhood on that Mountie, panting and strengthening her single functioning lung. In May 1980, returning home after her second spinal fusion which left her in a plaster cast from her chest to her hips and unable to swim, Sanders found a consolation prize in her living room: her first 10-speed bike, the model aptly named Dream Machine. Riding a bicycle was just about the only physical activity

approved by Sanders’ orthopedic surgeon. A bicycle is still Sanders’ dream ma-chine. The now 45-

year-old is cancer free and a two-time PMC cyclist. During PMC weekend, she was inspired by the riders, volunteers, and sup-porters around her committed to fighting cancer. “Throughout the ride, I thought of my mother who lost her life to esophageal cancer three years ago, and how she wanted to one day ride in the PMC,” said Sanders. “I also thought of that moment in middle school when my father told me I had had cancer as a baby. And my mind flipped through my vari-ous childhood bicycles. Now I realize that my parents gave me each of those bikes as I was growing up so that I would push my physical limits a little harder.” Sanders added, “I think they’d approve of what I’m doing with my bike these days.”

Father’s Day with an Extra Special Meaning

Jack Manning threw a perfect strike at Fenway Park. Haven’t heard Manning’s name in the Red Sox pitching line-up? That’s because he’s 16 years old. He and his father, Vincent, threw out the ceremonial first pitches—both of which landed in the catchers’ mitts—in front of thousands of fans at PMC at Fenway Park, an event that celebrated the Red Sox Foundation’s twelfth year as the PMC’s pre-senting sponsor. The Red Sox took on the Cleveland Indians that Father’s Day. Taking the mound before the game, and doing so in front of so many fathers and sons, was especially meaningful for the Mannings. Jack is a cancer survivor, amputee, and first-time PMC cyclist. In 2006, he was suc-cessfully treated for osteosarcoma at the Jimmy Fund Clinic. Since then, Jack has con-

tinued to serve as a pitcher on his baseball team and played football, basketball, and many other sports. The father-son duo rode from Sturbridge to Provincetown as part of Team Jack in what marked Vincent’s fifth PMC ride in honor of his son. “It was awesome throwing out the first pitch and also riding in the PMC this year with my Dad,” said Jack. “It was great riding with the team and show-ing that you can do anything you want after cancer. You just have to get up, get yourself back into sports, biking, or whatever you love, and just keep going.” Vincent said it was an honor to be a part of the celebration at Fenway. “It was a special day for our entire family and was topped only by PMC weekend itself,” he said. “We were so proud of

From Hat to Clipboard to Handlebars for Denise Singleton

Denise Singleton started out 11 years ago volunteering at the Brewster water stop. That same year, her brother and his wife, who had just completed treatments for an aggressive form of breast cancer, began riding in the PMC. For three years, she relished volunteer-ing but during those years, two more family members were diagnosed with cancer, and Denise wanted to do even more. Nervous to ride the PMC due to arthritis, she started the Westwood PMC Kids Ride in 2008, an event that has since raised more than $380,000.

As time passed, Denise has had more loved ones affected by cancer, including her neighbor, Maura Ryan, a mother with three young children, who is currently battling breast cancer. In 2012, Maura’s husband Keith start-ed Team Mo-mentum in honor of his wife. This year was supposed to be Maura’s first year pedaling on the team, but a training accident sidelined her. With Maura unable to ride Densie decided it was time to get in the saddle herself. This August, Denise, the newest member of Team Moo-memtum,

conquered the one-day, 82-mile PMC route from Wellesley to Bourne. In a way, like many of the Westwood PMC Kids Ride cyclists, Denise took off her training wheels. “I felt the same pride, passion, and power in each of my roles, as PMC volunteer, PMC Kids Ride Coordinator, and PMC rider,” says Denise. “The PMC organization truly values everyone who takes part and recognizes that we all are so critical to its mission.”

New PMC App Developed by PMC Alumni Tech CEOs Breadcrumbs for tracking. That’s the basis of a mobile app that debuted on iOS devices for PMC 2014 and will be available by 2015 on Android devices. “Through its breadcrumb technology, the app keeps tabs on riders periodically and shuts itself off between pings, allowing riders to be tracked for an entire day without draining their phones’ batteries,” says PMC Vice President of Operations, David Hellman. “No other app makes it possible for a rider to be tracked for a century ride without a battery refill.” Hellman worked closely with app developers Paul English and Ben Fishman. English, a three-year PMC rider, is the founder and former CEO of Kayak.com and is now the CEO of Blade, Boston’s hottest technology incubator. Fischman, a seven-year rider, is the founder and former CEO of Rue La La and the founder of Launch, a platform to create new innovative ecommerce businesses. With first-hand knowledge of the PMC, English and Fischman came together in Spring 2014 to develop the mobile app uniquely suited to PMC weekend and year-round needs. The PMC app enabled riders to track each other and their own progress over the course of the two-day, 190-mile event. Riders’ friends and supporters could download the app to stay updated on riders’ progress. The location data transmitted by the app supported the “Call for Help” feature riders used to seek immediate aid. The PMC Command Center used the data to quickly send riders assistance in the case of accidents, mechanical problems, or those who mistakenly strayed from the route. Riders training for PMC 2015 will also be able to map routes and save them through the PMC mobile app. As the app continues to develop, it will also allow riders register to ride and manage personal fundraising in 2015. Riders will be able to email or text contacts directly from their mobile devices to seek donations, track gifts, or send thank-you messages. The app will provide the same data about sponsors that is available through the PMC website. This powerful tool for PMC riders is available in the App Store.

Plenty of people might want to know Dan Salon’s secret. He’s 10 years old and this year raised more than $9,000 for the Hingham-South Shore PMC Kids Ride, which he rode in June. He did it for his mom, Karen, who was diagnosed in January 2014 with colon can-cer that has since spread to her liver. Karen,

who has two rounds of chemo to go, couldn’t be prouder of Dan and her other children, Ethan, 15, and Jane, 12, who, in addition to raising money and awareness for cancer research, pitch in around the house to prepare meals and do other chores. Incredibly, Dan already has eight years of fundraising experience. When he was 2, he rode in the inaugural Hingham-South Shore PMC Kids Ride alongside his siblings. Karen can’t remember a time when their family hasn’t been cognizant of the need to support Dana-Farber, as the leading cancer center in their area. “We’ve all been, or will be, touched by cancer in some way, whether it’s affected our friends, parents, or people in our commu-nity,” she said. “When the PMC began the PMC Kids Rides Program, we jumped at the chance for our kids to help in a tangible way.” The kids hosted a lemonade stand in their neighborhood for their PMC fundraising cam-paign. This year Dan added social media to his fundraising strategy, though since he doesn’t yet have permission to have his own Facebook page, he asked his parents to promote his ride through their Facebook pages. That netted an outpouring of financial and moral support. For his efforts, Dan was honored as a Red Sox Little Star (right on the field!) during PMC Day at Fenway Park and announced players as they came to bat. One day last summer, Dan spent a day at Dana-Farber with his mother while she received chemotherapy. He toured the building and met the doctors, nurses, and social workers who collaborate to help treat Karen. Dan learned about how the money he raises funds research. He was downright impressed with the facility and proud that he was able to contribute, commenting on his profile page “We are lucky that my mom has the best doctors in the world and that we live so close to Dana-Farber, which is one of the best cancer centers in the country.”

PMC Head Staff Profile: Matt McGuiness, Old-Timer at 33

PMC weekend is seemingly etched as long and deeply as hiero-glyphics on Matt McGuiness’s calendar. Matt, at 33 years old, is the youngest, longest-standing PMC volunteer, with 22 consecutive years of PMC service. The family tradition started when McGuiness was 11 years old. His father Stephen became involved as a rider and a volunteer at the Sturbridge site to honor a neighbor with cancer. That year, he enlisted Matt to help load and unload trucks. Matt did such a good job that next year he was promoted to Chief Garbage Collector, a title he wore proudly. Years later, when Matt was asked to co-coordinate the Sturbridge site with Sue Brogan, he made sure he divided the clean-up responsi-bilities among all the site’s volunteers. “No one wants to have his head in the trash the whole weekend,” said Matt. Now he oversees every-thing from food to logistics, working with the staff of the Sturbridge Host Hotel for months leading up to PMC weekend and heading up 15 crew leaders. With this many years under his belt and at a site as large as Stur-bridge, he’s amassed some amusing anecdotes along the way: like the time there were no plastic forks on site and nearly 4,000 riders were expected for dinner. Or the time the PMC was short bedding for 60 riders who were spending the night at a nearby college. On both occa-sions, volunteers saved the day culling items from near and far. There was also the year the parking lot flooded in heavy rains, nearly wash-ing away cars. Once again volunteers stepped in to assist riders wade through knee deep water to save their belongings. “The best kind of volunteers are the ones who are flexible since it’s hard to anticipate what will happen, even though the PMC is a well-oiled machine,” says McGuiness. With Matt’s PMC track record, it shouldn’t be surprising that many of his most flexible volunteers happen to be family. Matt’s mother, wife, and in-laws all have been a part of the Sturbridge volunteer crew. His mother heads up the “avoiding potential disasters” department, he says, only half joking. Matt can’t seem to stop giving his time even after the Sturbridge site is packed up and the cyclists have hit the road. After three years of cycling on

PMC Sunday in 2010 through 2012, he took on the additional role of site coordinator at Patriot Place to make further improvements so that one-day riders with varying abilities are assured of a great experience. Through medical school, internships and residencies, Matt never missed a PMC weekend. This past July, Matt and his wife Michelle welcomed their first child, Lauren, just before the PMC. He is now a cardiologist in UMass Memorial Medical Center’s cardio-oncology department, where he works with cancer patients who have experi-enced heart damage as a result of chemotherapy treatments. “The PMC is an easy event to love,” says McGuiness. “Everyone is so committed to making a difference in the lives of cancer patients. And that commitment comes full circle for me now that treating cancer patients is part of my job. Both my job and volunteer work are so rewarding for me.” In 2015, the third generation McGuinness, Lauren, will be a “vol-unteer” cheerleader at the Sturbridge start alongside her parents. “She has already mastered waking up early to cheer,” says Matt.

A Family Team Rides to Stay Connected to a Mom and Wife Lost to Cancer The love and respect that Mike Maher, 47, has for his late wife, Jen, drives his momentum in the PMC. Jen rode in the annual bike-a-thon for four years before being sidelined in 2004 with a diagnosis of naso-pharyngeal carcinoma, a rare form of head and neck cancer. That year, Maher took her place and rode the 190-mile route in her honor. “When my wife asked me to ride in her place, I was nervous as I hadn’t been on a bike since I was in middle school and had baseball cards on my spokes,” says Mike. “But a good friend and teammate lent me a bicycle and took me on training rides. My first PMC was very emotional, especially when my wife met me at one of our local water stops.” After enduring one year of surgeries, chemotherapy, radiation, and clinical trials, Jen got back on her bicycle and joined her husband in the 2005 PMC. During the ride, she experienced leg-stopping back pain. Tests revealed that her cancer had resurfaced through her spine and metastasized to her liver. In March 2007, she lost her life to the dis-ease. Three months later, Mike was diagnosed with stage II melanoma. His cancer was caught in an early stage, and after undergoing surgery to remove the cancerous lymph nodes under his arm, he was declared cancer-free. This August, Mike led Team JEN-STRONG, a group of eight friends and family members who rode the PMC in Jen’s memory. What made

this year’s ride extra special was the newest member of the team, Mike and Jen’s son, Connor, 19, who pedaled the 163-mile route as a first-time rider. To date, Mike has raised more than $136,000 and has been named a PMC Heavy Hitter each of the 10 years he has par-ticipated. As a Living Proof cyclist, a PMC rider, and a cancer survivor, Mike’s goal is to give back to Dana-Farber for treating his wife. And he plans to recruit all three of his children to ride and raise money as a tribute to their mother. “I am so proud of Connor for his many accomplishments in life, but crossing the finish line with him by my side brought me back to the day he was born when I held him for the first time and felt pure joy,” says Mike. “The PMC is now in my family’s blood. I wear an article of PMC clothing almost every day. Our annual fundraising and training is a time when we feel Jen’s strength and spirit within us even more powerfully.”

A Truly Special MA License Plate

The Massachusetts Registry of Motor Vehicles (RMV) is about to begin pro-duction of the PMC special license plates. More than 1,500 riders, vol-unteers and donors signed up for the program, which costs $40 every 4 years on top of the normal vehicle registration fee. Once 3,000 plates are in circulation the PMC will receive $60,000 every four years. Closer by the Mile is integral to the PMC identity and is part of the badge, which decorates the plate. Share the Road is a message to be taken to heart by all cyclists and vehicle operators. Bike shops across the state have taken up this pledge and shared the special plate program with their constituency.

Jonathan Boyer, the first American to ride the Tour de France, formed Team Rwanda after his move to Rwanda in 2007. The team is the subject of of the documentary Rising from the Ashes, narrated by Forrest Whitaker and has received international attention for their ability to overcome adversity. Composed of roughly 18 men and women, Team Rwanda competes in Africa, North America, and Europe with the mission of unifying African countries through the sport of cycling. Through a PMC rider, the PMC donated past ride jerseys for the team to wear while training.

A Diagnosis, A Career, A Patient, A Ride Written by Magie Loucks

When I was 27 and in my last year of graduate school studying to be a nurse practitioner, I found a lump in my left breast. Many diag-nostic tests later came the call from my primary care physician. I don’t remember much from that call other than the worry in my doctor’s voice and her murmured phrase, “the lump is malignant.” I felt as though the floor fell out from beneath me. The fear of the unknown was crippling beyond words. I was unsure how to take another step forward. The only thing I was certain about was that I had to go to Dana-Farber. In my opinion, it was my only option. Given my age and diagnosis, a friend helped guide me to Dr. Ann Partridge, truly a world leader and pioneer in the fight against breast cancer in young women. Partridge leads the only Program for Young Women with Breast Cancer in the country. Four days after I completed my first leg of treatment, including 12 straight weeks of chemo, I graduated from UMass Medical School as a nurse practitioner. Prior to my diagnosis I was unsure of what type of nurse practitioner I should be. The medical care that I received at Dana-Farber by Dr. Partridge and her team had such a profound influ-ence on me that I wanted to emulate it with patients I would be caring for. I was fortunate enough to be hired as a nurse practitioner at Dana-Farber’s Center for Sarcoma and Bone Oncology, and worked on the same floor where I was treated. I learned about the PMC through a very special and dear patient of mine, Erica Kaitz, who was the special events coordinator for the PMC. Knowing she had sparked my interest, Erica selflessly allowed me to borrow her bike to “test it out.” Not only did Erica’s act of kindness get me completely hooked on the sport of cycling, but her bravery, strength, and unparalleled graciousness inspired me and my husband Drew to ride the PMC this year. Erica’s undeniably kind and caring spirit instilled me with an implausible amount of energy and vigor to ride 164 miles from Wellesley to Provincetown. There are no words to properly articulate the incredible feeling I experienced while I rode in my first PMC. If it is any indication, I couldn’t stop smiling the entire way, despite the bitterly cold, rainy weather. The PMC is unlike anything I have ever experienced before and I am forever grateful to Erica for getting me involved in this amazing event.

“Who are we riding for?” That one question led Shawmut Design and Construction to create a dozen or so inspiring signs—some held by support-ers, some posted along the route lead-ing up to the Brewster Waterstop. So who was Team Shawmut riding for? They were definitely riding for fel-low employees and their families, but as President and COO Les Hiscoe said, “We were also riding for thousands of people we don’t know. Their families. And their friends.” With that in mind, Shawmut enlisted volunteers and got to work. The result was even better than they had hoped. “Riders were actually thanking us,” said Shawmut volunteer and sign-holder Tim Kelley. “We were really moved by that. I mean, we should be thanking them!”

Harpoon Creates PMC Beer to Hoist Fundraising Even More Quenching PMCers’ Thirsts for a Decade: That’s a watchword of Harpoon, the official beer of the Pan-Mass Challenge. The company supplies beer for the PMC Heavy Hitter event, the PMC Check Presentation, and—try lifting 180 kegs—during PMC weekend at the Sturbridge Host Hotel, Mass Maritime Academy, and Provincetown Inn. PMC Founder Billy Starr joined Harpoon staffers and veteran PMC cyclists Jamie Schier and Jamie Rudavsky in early July to brew Harpoon PMC beer, a light beer with a hint of lemon zest, pineapple and coriander, on Harpoon Brewery’s brew deck. The beer was sold over three weeks in Harpoon’s Beer Hall in Boston, with $1 per pint benefitting the PMC, raising $1,575. Sixty kegs of that Harpoon PMC beer were made available to PMC riders at the Mass Maritime Academy. “There’s a huge correlation between cycling and beer. Plenty of people like both,” says Schier. “Both are fun and social endeavors that are made better by sharing with friends.” Schier and Rudavsky ride the PMC as part of Team FORZA-G. Schier rides in honor of three friends lost to cancer. Rudavsky rides in honor of his father, a two-time cancer survivor.

“There is nothing better than an ice cold Harpoon after a long day in the saddle,” Rudavsky said, “and knowing that everyone who rides the PMC can enjoy a Harpoon, too, puts a smile on my face. It feels great to be a part of something that each rider will be able to enjoy after arriving in Bourne.”

Former PMC Pedal Partner Pays it Forward

Ashley Cunningham’s focus went from surviving the academic and social challenges of high school to undergoing a year of multiple spinal taps, bone marrow aspirations, and chemotherapy treatments at Dana-Farber Cancer Institute’s Jimmy Fund Clinic. At 15, Ashley was diagnosed with non-Hodgkin’s lymphoma. She gained comfort from Team Bain Capital, a group of PMC cyclists who developed a relation-ship with her family through the PMC Pedal Partner Program and who rode the annual bike-a-thon in her honor. The PMC Pedal Partner Program, now in its 14th year, matches Jimmy Fund Clinic patients with teams of PMC cyclists. “When I was a PMC Pedal Partner, I met Heidi Flood, one of the cyclists from Team Bain Capital, at the PMC Pedal Partner Party, and she was incredible, our personalities just clicked,” says Ashley. “Heidi was no stranger to cancer as her mother battled the disease and she was young enough to relate to me as a young woman. That summer, a hand-ful of cyclists from the team met me at the PMC Pedal Partner Tent in Lakeville. I was grateful that total strangers would dedicate 190-miles to me as a way to benefit the Jimmy Fund. This spring, I had lunch with Heidi, who also came to my high school graduation party. It is so amazing to know that these strangers had my back through-out treatments and are continuing to sup-port me as a survivor.” Ashley, now a 19-year-old rising sopho-more at Boston University, has been in remission for two years. She shared her story of resilience and recovery with current

Pedal Partners and their teams at the annual PMC Pedal Partner Party at Fenway Park this past spring. During PMC weekend, Ashley and her mother, Fran, volunteered at the PMC Pedal Partner tent at the Lakeville water stop. There Ashley spent time with Team Bain Capital’s two-time PMC Pedal Partner, Ruby Gan, 4, who is currently in treatment for acute lymphoblastic leukemia at the Jimmy Fund Clinic. Ashley chatted with the Gan family about her studies and college experience thus far under the superhero themed-tent as they eagerly watched for members of Team Bain Capital, including Ashley’s father, John, to stop by. Ashley plans to serve as a resource to current and future PMC Pedal Partners and their families. She also aspires to become a chief oncology nurse, and is majoring in health science.

PMC Pedal PartnersThe Pedal Partner program continues to inspire PMC riders to train, ride and fundraise. Seventy Pedal Partners, oncology patients at Dana-Farber’s Jimmy Fund Clinic, were matched with PMC teams and Kids Rides who rode in their honor. The Boston Globe has sponsored the

PMC since 2011, donating 36 ad place-ments totaling over $600,000 of in-kind media value. With a circulation of up to 382,000, The Globe has helped the PMC recruit new riders, volunteers, and support for fundraising. Rides and volunteers often men-tion seeing the ads, and the PMC is frequently asked how riders are chosen and who they are. “It’s that certain joie de vivre” said David Hellman, PMC senior VP of operations. For many years, doctors were unable to provide the most basic information about cancer to their patients—why do individuals get cancer. The advent of human genome sequencing opened the doors to cancer genome sequencing, which can now be performed for indi-vidual patients bringing us ever closer to precision, personalized cancer care. The premise of this promise is to deploy next-generation cancer therapeutics innovated to target the deranged (or mutated) gene that underlies each individual patient’s cancer. For sure, this strategy has already changed the lives of patients with types of leukemia and a select number of aggressive solid tumors. But the news is not all good. Only in the dawn of cancer genome sequencing, we are starting to learn the scope of the challenge ahead. There are, to date, more than 1 million alterations observed in cancer genomes, found in more than 20,000 genes—which is all of them. While many of these changes are bystander effects, at least 140 and likely as many as 500 of these changes contribute to the aggressive behavior of cancer. To deliver on this promise of precision cancer medicine, then, we may need a hundred or more new targeted thera-peutics. Despite decades of collaborative research between academia, the government and pharma, there are just 15 genes for which a targeted cancer drug is presently approved. This suggested to us at Dana-Farber that we may not only need new breakthrough medicines to fight this disease, but we may need to rethink the manner by which drugs are discovered and developed. In 2003, Dr. Stanley Korsmeyer and President Edward Benz became tired of sitting on their hands, waiting for new drugs to arrive. They set an ambitious agenda, to bring chemistry—the science of drug discovery—to the leading cancer biology facility in the world. You see, Dana-Farber had tested more drugs than almost any other center in the word, but never developed one of its own. This experiment would be costly, requiring the recruitment of faculty trained in chemi-cal synthesis and the reconstruction of research labs to accommodate the specialized needs of this science. There is no grant for this type of project. The federal government has no mechanism for building out the laboratories that were needed. Enter the PMC.

Where the Money GoesWritten by Dr. Jay Bradner

So I want to tell you where the money goes. They hired three chemists, and I am deeply honored to be one of them. After completing my training as a cancer doctor, unsatisfied with the perfor-mance and creativity of modern cancer drugs, I returned to retrain in chemistry. This science applied to cancer research, has the power of revealing new insights into these diseases while offering the immediate promise of therapeutic intervention. It is early days for our program, but already we are sensing the power of bringing chemistry into prox-imity with biology. In only eight years, seed funding to three labs have generated 300 scholarly publications, 49 new grants that recruited 58 leading scientists, 50 prototype drugs distributed broadly and openly to the scientific community, and 6 new drugs presently in human clini-cal trials. Most importantly, we are already observing these medicines improve the outcomes for our patients. Ambitious projects, such as the development of a drug discovery capability, require unrestricted funding that allow the Farber to throw down when needed and when uncertain. In January, we will expand this program to a state-of-the-art discovery chemistry facility called the Longwood Center. We will recruit eight new professors, establishing the strongest faculty in cancer drug discovery anyplace in the world. This is why I ride. I believe we are on the verge of changing the course of history for cancer patients, but we cannot as scientists defeat cancer in the isolation of our laboratories. Through this ride, this region, this country, we rally around the science and care at Dana-Farber in a way I have never seen before. I ride in awe of your convic-tion, and I thank you personally for your confidence in our ideas.

A Decade of Riding in the Pan-Mass Challenge. A Lifetime of Lessons LearnedWritten by PMC rider Nicole Barucci

This year marked my 10th consecutive Pan-Mass Challenge. I am proud of that. Not proud that I have ridden thousands of miles. Not proud of the training endured. Not proud of spinning, late, early, after dark, after work just to get a ride in. The PMC is not about me. And that’s the beauty of it. In a decade of riding, I have learned more about the human spirit than I ever thought possible. I have learned that people are at their very best when they’re doing something selfless. Strangers. Clapping and cheering. And thanking me. Over and over. In the pouring rain. In the extreme heat. Thank you. I am here, because of you. To hear those words, and see their faces as they thank you with their whole hearts. Sincere. Pure. It

continues to overwhelm me. Year after year. I believe them. We are making a difference. A real, life-saving, cancer-fighting difference. They are Living Proof. I have learned that some of the most generous of people are the ones you barely know, but know you. I have learned that PMC volunteers have a tougher job than any cyclist does. They are kind, patient indi-viduals who smile with their hearts. Angels. All of them. I have learned that the smallest of gestures requiring the most minimal of time are the most profound. A ribbon honoring a loved one’s fight. A quick note saying, “I will be thinking of you this weekend” means the world to people. Find the time. You are not that busy. It’s worth it. I am proud to be part of the PMC com-
