A method will be developed to carve out grooves in the subchondral bone to allow gentle bending of these allografts for the purpose of increasing their curvature and matching it to that of the natural thumb CMC articular layers.

Description: This grant will study the dynamic osmotic loading response of cultured chondrocytes using a custom microfluidics device. The role of cell size change and osmotic load magnitude/frequency on the intracellular calcium signaling response and subsequent cytoskeletal rearrangement and aggrecan gene expression of chondrocytes will be assessed.

Description: This project merges microbubble technology to create micropores in hydrogel scaffolds that can aid nutrient access in growing engineered tissues by shortening the diffusion path length. Gas-filled microbubbles can be triggered to be filled by fluid.

31. Principal Investigator: NIH: NIAMS 2R01AR46568-12: Physiologic loading for cartilage tissue

engineering; approximate yearly direct cost: $225,000, 9/1/09-8/31/14 (total cost for entire granting period: ~$2.38 million)- 20% yearly effort. Description: This COMPETING RENEWAL continues our investigation of the use of applied physiologic loading in a custom bioreactor to grow functional articular cartilage. Engineered constructs preconditioned with loading or free-swelling conditions will be implanted into a canine knee defect model with collaborators at the University of Missouri-Columbia (JL Cook, subcontract PI) to test the hypothesis that constructs cultured with physiologic loading will lead to better tissue repair. Complementary theoretical modeling of the biomechanical conditions at the defect site will be performed and (strain fields) validated using cadavers.
