A STAR-STUDDED ARRAY OF CELEBRITIES joined Grammy-nominated songwriter andG&P Co-Founder Denise Rich at the New York Marriott Marquis for the fourth AngelBall gala, helping to raise $3.3 million for The G&P Foundation for Cancer Research.The black-tie benefit, which featured a cocktail reception, dinner and live auction,recognized four honorees for their outstanding contributions as humanitarians andphilanthropic leaders: singer/songwriter Patti LaBelle; Russell Simmons, Chairmanand CEO of Rush Communications; Kimora Lee Simmons, Creative Director, BabyPhat; and GLAMOUR Magazine Editor-in-Chief Cindi Leive.

The triumph of the night’s fundraising efforts was highlighted by a $1 milliondonation from the royal family of Abu Dhabi. Due to the great success of this event,The G&P Foundation will fund seven new Medical Research Awards in 2006 with theproceeds from Angel Ball 2005.

Legendary vocalist Patti LaBelle segued from her acceptance speech into a surprise,heartfelt rendition of “Somewhere Over the Rainbow”, and was joined by StevieWonder at the piano, Natalie Cole and Wonder’s daughter, Aisha Morris. The nightculminated with all of the performers uniting onstage to sing the Martha Reevesclassic “Dancin’ in the Streets.”

The Angel Ball 2005 also featured performances by Ruben Studdard, Nelly, LeelaJames, BeBe Winans, Miri Ben-Ari and Shawn King. Other celebrities participated bydonating their time and talents as presenters including Star Jones Reynolds, KellyRipa, Lorraine Bracco, Joy Behar and Montel Williams (both served as the evening’sauctioneers), Jamie-Lynn Sigler, Ivana Trump, Joan Collins, JoJo, Debra Lee andFelicia Taylor, among others.

The Angel Ball is held biennially in memory of Gabrielle Rich Aouad (1969 - 1996).Angel Ball 2005 was sponsored by Donald Toresco of Toresco Enterprises, Inc.,Condé Nast Publications and PhRMA.

In this third installment, we are proud tointroduce you to Janet Davison Rowley, M. D.of The University of Chicago. Dr. Rowley isthe Blum-Riese Distinguished ServiceProfessor in the Departments of Medicine,in Molecular Genetics and Cell Biology andin Human Genetics. She received hermedical degree from the University ofChicago in 1948. She joined the researchfaculty at the University of Chicago in 1962and became a Professor in 1977. Dr. Rowleyhas contributed significantly to advances inunderstanding of genetic changes in cancer.Her discoveries have revolutionized the viewof hematologists/oncologists and cancerbiologists regarding the critical importanceof recurring chromosome abnormalities incancer cells.

She has published over 400 papers and bookchapters describing her research inleukemia and related areas and has receivednumerous awards including Dameshek Prize(1982), Medal of Honor (ACS, 1996), and morerecently the Dorothy P. Landon Award(2005). She is a member of the NationalAcademy of Sciences (1984), Institute ofMedicine (1985), American Academy of Artsand Sciences (1991), and AmericanPhilosophical Society (1993). She hasreceived nine honorary degrees includingone from Oxford in June 2000 and fromLund in 2003.

JANET DAVISON ROWLEY, M. D.

“I should begin by saying how important Ithink the work of the Foundation is,especially today as budgets for science arebeing constrained by other federalrequirements. The goal of The G&PFoundation namely to fund young scientistsand physicians and others involved in thecare of patients with cancer is more neededthan ever. Young scientists have a very hardtime breaking into the system and gettingfunding for starting their laboratories. I wasvery fortunate back in 1962 when I began inresearch to be part of a project that wasfunded by the national government so Ididn’t have to apply for grants in the firstfew years.

The project that I have been involved infor more than thirty years is the study ofgenetic changes in leukemia cells andlymphomas. The tools I used to studythese genetic changes in the malignantcells were chromosomes. The normalhuman cell has 46 chromosomes of partic-ular sizes and shapes. What has becomeapparent over the course of my studies isthat many leukemia cellshave altered chromo-some numbers (too manyor too few) or they havealtered chromosome sizesand shapes due to breaksin chromosomes andrearrangements.

I was very fortunate in1972 to be the first toidentify the chromo-somes involved in two ofthese re-arrangements,called “translocations”.In cancer and especiallyleukemia and lymphoma,chromosomes are brokenin specific places, andtheir ends are swappedso that different size pieces of the chro-mosomes are exchanged. This leads toidentifiable changes in the chromosomeswhen you look at them under the micro-scope.

The most well known translocation I dis-covered was one involving chromosomes9 and 22. It had been known since 1960that a particular form of leukemia calledChronic Myeloid Leukemia (CML) had aunique small chromosome. Because itwas discovered in the city of Philadelphiait was called the Philadelphia or the Phchromosome. With the staining tech-niques that were used at the time, chro-mosomes all stained uniformly. Theycould identify this very small markerchromosome but how it came to be wasn’tclear. I applied a new technique thatshowed that each chromosome pair had aunique pattern of alternating dark andlight stripes to study this mystery markerchromosome. I could see that in fact thePh chromosome was actually a transloca-tion where a large piece of chromosome22 went to the end of chromosome 9 and asmaller piece of chromosome 9 went tochromosome 22 giving rise to this smallPh chromosome.

The importance of this is that other labo-ratories analyzed the DNA at the sites ofthe chromosome breaks and discoveredthat the gene on chromosome 9 was theAbelson gene, which had already beenidentified in a mouse leukemia. The con-sequence of the translocation was tobreak within the gene and to move part ofthe Abelson gene to chromosome 22, next

to a gene called BCR thatis also broken. So youend up with a new,unique fusion gene onthe Ph chromosome,which consists of part ofthe normal BCR gene on22, fused to the back half,if you will, of theAbelson gene, which isnormally on chromo-some 9.

This fusion leads to theactivation of the Abelsongene whose function isas a tyrosine kinase. Thetyrosine kinases act toincrease cell growth.The fusion then leads

these cells to grow not necessarily morerapidly but it keeps them from stoppinggrowth when there is sufficient number ofwhite blood cells. This, in turn, withother genetic changes whose identity isunknown at the present time, leads thecell to become a leukemic cell.

This discovery has turned out to beextremely important because others, mostespecially Brian Druker at Oregon Health& Science University, working with achemist at Ciba, identified the compoundknown as STI 571 that inhibited thegrowth of cells with this abnormalAbelson fusion protein. This drug wasthen named Gleevec, now Imatinib, whichhas transformed the treatment of patientswho have CML.

The challenge for the future is to identifythe same kind of drug to treat patients whohave acute forms of leukemia, to treat themmore effectively. This is especially urgent forpatients who have Acute MyelogenousLeukemia (AML) who are older, i.e. over theage of 60, because the outcome oftreatment in these patients is still dismal.So, there is need for continued research tofind additional drugs to treat these otherforms of leukemia.”
