Air Sampling for Total Spore Counts

AIR-O-CELL • TheAir-O-Cell™cassette(ProductID8715301B)isasingle-usesamplingdevice designed for the rapid collection and analysis of a wide range of airborne particles. These include fungal spores, pollen, insect parts, skin cell fragments, fibers, and other inorganic particulates. • Thecassetteisdesignedtooperateataflowrateof15LPM.Lowerflowrates may result in a collection loss of some spores and the accumulation of others in a non-uniform manner. Therefore, it is critical to run the sampling pumps at the manufacturer’srecommendedairflowrate.

Benefits • Usefulforinitialsitetesting,especiallyiffungalgrowthisnotvisible. • Quickandsimpleprocedure. • Fastturnaroundtimesavailable. • Lowchanceofsamplecontamination. • Necessaryfordeterminingallergicmoldsporepotential. Moldsporescancauseallergieswhethertheyareviable ornon-viable

Disadvantages • Fungicannotbeidentifiedtospecieswiththismethod. • Duetothesimilaritiesinsporemorphology,somesporeswillbe grouped together, i.e., Aspergillus sp. and Penicillium sp. • Sporeviabilitycannotbeassessed.

Sampling Procedure 1. Priortosampling,calibrateyourpumpto15litersperminuteviaarotameter.Itisalsorecommendedthat therotameterorsamplingpumpbeperiodicallycalibratedtoaNISTprimarystandard.IfusingtheZefon Bio-Pump(ProductID8706002)usethespecificallydesignedAir-O-Cellflowindicator,aspeciallydesigned bubbletubeNISTPrimarycalibrator(ProductID8704200).Itcannotbeusedwithanyothertypeofpump. 2. Removeandretaintapesealcoveringinletandoutletonthecassette. 3. Attachtheoutlet(roundhole)toastandard1/2”PVCtubing(forusewithhighvolumepumpsonly). 4. Start the sampling pump and sample for an appropriate period of time (see recommendations below). 5. RemoveAir-O-Cellfromtubingandresealwiththeoriginalseals.Labelsample. 6. CompleteanEMSLChainofCustody(COC),availableatwww.emsl.com,detailingclientnameand information,projectnameornumber,sample#,descriptionofarea,andvolumeofaircollected. 7. Toreduceshippingdamage,itisrecommendedthattheAir-O-Cellbeplacedinacorrugatedboxwith paddingtoensuresafearrivalatthelaboratory.

Sampling Duration • Thesamplingtimeisdependentonthedensityofparticulateintheenvironment.Itisimportantnotto overloadthesample,otherwiseitwillbeimpossibletoaccuratelycountthetypesofspores,pollenorother particulates that are present. The following list represents typical sample times to attain a sharply defined trace with good dispersion of the spores: 1.Clean“office”oroutdoors(novisibledust)=10minutes 2.Indoorenvironment,highactivity&personnel=5minutes 3.Indoorenvironment,drywallrenovationorheavyindustrialdust=1minute.

Quality Control Recommendations • Aneffectiveinterpretationisbasedonthecomparisonofindoorandoutdoorsamples.Outdoorsampleswill help demonstrate whether spore amplification is occurring indoors. • Obtainacontrolsamplefromanon-complaintareaforcomparison. • Sendingablankcassetteforanalysisperprojectisagoodpractice. • Flowrateiscriticalforaccurateresults.Remembertocalibrateandrecalibratethepumppriortoallsampling. Tech Tip: Basic Mold Cleanup

The key to mold control is moisture control. It is important to dry water damaged areas and items within 24-48hourstopreventmoldgrowth.Ifmoldisaprobleminyourhome,cleanupthemoldandgetridofthe excess water or moisture. Fix leaky plumbing or other sources of water. Wash mold off hard surfaces with detergent and water, and dry completely. CulturableAirSampling(FungiorBacteria)

ParticleImpactors(Andersen-typeSamplers) • ThismethodofairsamplinginvolvesdrawingameasuredvolumeofairoverculturemediainPetridishes. The Petri dishes are incubated in the laboratory so the organisms impacted on the plate can grow. The fungi orbacteriaarecountedandidentified.ThismethodcommonlyusesanAndersenN-6typeimpactor(e.g.EMSL VP-400MicrobialSamplerProductID8709001).DifferentagarplatesareavailablefromEMSLAnalytical, Inc., depending on the types of fungi or bacteria to be identified.

Benefits • Fungalculturescandeterminewhetherthefungusisviable(alive),andallowsforgenusandspecies identification. • Bacterialculturesprovideenumerationandidentificationofculturablebacteriapresentintheair.

Disadvantages • Culturestake6-10daysforthemicroorganismstogrowandbeidentified. • Sincemostenvironmentalsamplescontainalargenumberoforganisms,eachhastocompetewithothersto grow on the media. As a result, fungi and bacteria present in the air may not be as well represented in culture. • Somemicrobesdonotgrowwelloratallontheculturemedia. • Somemicroorganismsareunabletobeidentified,astheyfailtoproducekeycharacteristicssuchassporesor they may not be described in the scientific literature.

HowtoHandleMicrobiologicalMedia(Agarplates) • Agarplatesmustbekeptrefrigeratedoronfreezerpacksuntilreadytouse. • Theplatesmustbeallowedtowarmuptoroomtemperaturebeforetakingasample(approx.15minutes). • Donotremovethelidfromtheplateatanytimeexceptduringsampling. • SealthelidtotheplateaftersamplecollectionwithParafilmortape. • TheplatesmustbeshippedbacktoEMSLwithfreezerpacksbyOVERNIGHTPRIORITY. Refreezeandreusetheoriginalfreezerpack(thistypeoffreezerpackisstablefor24hours). • Adequatepackingmaterialmustbesenttoprotecttheplates. • Platesmustnotcomeintodirectcontactwiththefreezerpack,asthemediamayfreeze,invalidatingthetests. • IfthereisanydelayinsendingtheagarplatestoEMSL,theyshouldberefrigerateduntilreadyfor overnightdelivery.

Recommendations • Wearlatexornitrileglovesduringsampling. • Use70%isopropylalcoholtodisinfectsamplingdevicebetweeneachsample. • PlacePetridishlidinacleanbagduringsamplingtoreduceanycrosscontamination. • Includeoutsidesamplesandafieldblankforcontrol.

Sampling Procedure 1. Allow agar plates to reach room temperature before use. 2. Attachoneendoftubingtotheintakeofthevacuumpumpandtheotherendtotheinletofthesampler. 3. Calibratetheflowrateofthevacuumpump: a.PlaceanuncoveredPetridishintosampler(Donotsubmitthisdishasasample,discardaftercalibration). b.Turnonpumpandadjustflowuntiltherotameterisat28.3LPM (flowrateisreadfromthemiddleofbearingontherotameter). c.Removestopperandrotameterpriortosampling. 4. Wipeallexposedsurfacesofsamplerwitha70%isopropylalcoholpadandallowtoairdry. 5. Place the agar plate on the sampler base so that the Petri dish rests on the three raised metal pins. 6. RemovethecoverofthePetridishandplaceintoacleansamplebagtominimizecontamination (availableuponrequest). 7. Assemble the jet classification stage on the sampler and secure the inlet cone with the three attached clips. 8. Settimertoappropriatetimedependingonenvironmentalconditions(samplingtimeisusually between 2-5 minutes). 9. Turn on the pump and start the timer simultaneously. 10. When the time is up, turn off the pump and disassemble sampler and placecoverbackontoagarplate. 11. Secure lid onto Petri dish with masking tape or Parafilm (avoidusingelectrical,packing,transparentandducttape). 12. Write the sample number on the bottom of the Petri dish. 13. Record all appropriate information on the Chain of Custody. 14. Return samples with an ice pack to EMSL Analytical for analysis.

Tech Tip: Canmoldcausehealthproblems?

Molds are usually not a problem indoors, unless mold spores land on a wet or damp spot and begin grow-ing.Moldshavethepotentialtocausehealthproblems.Moldsproduceallergens(substancesthatcancauseallergic reactions), irritants, and in some cases, potentially toxic substances (mycotoxins). Inhaling or touch-ingmoldormoldsporesmaycauseallergicreactionsinsensitiveindividuals.Allergicresponsesincludehayfever-typesymptoms,suchassneezing,runnynose,redeyes,andskinrash(dermatitis).Allergicreactionsto mold are common. They can be immediate or delayed. Molds can also cause asthma attacks in people with asthma who are allergic to mold. In addition, mold exposure can irritate the eyes, skin, nose, throat, and lungs of both mold-allergic and non-allergic people. Symptoms other than the allergic and irritant types are not commonly reported as a result of inhaling mold. Surface Sampling

DirectExamination(TapeLift,Bulk,Swab) • Adirectexamallowsfortherapiddeterminationofthepresenceoffungal spores as well as identifies the types of fungi. • Directexaminationsshouldonlybeusedtosamplevisiblemoldgrowthin acontaminatedareasincemostsurfaceswillhaveadepositoffungalspores thatarenormallypresentintheenvironment.

Benefits • Thedirectexamisinexpensiveandcanbeperformedquickly. • Ausefultestfordeterminingifthereismoldamplification. • Directsamplingmayrevealindoorreservoirsofsporesthat havenotyetbecomeairborne.

Disadvantages • Areasoffungalgrowthareoftensmallandscattered,sotheymaynotallbepickedup.Choosingmultiple samplinglocationswillhelpovercomethisproblem. • Healthproblemsrelatedtoindoormicrobialgrowtharegenerallycausedbytheinhalationofsubstantial numbersofairbornespores,sometimesoveralongperiodoftime.Thepresenceofbiologicalmaterialson a particular surface may not be a direct indication of what is in the air. • Thismethoddetectsbothviableandnon-viablesporesbutcannotdistinguishbetweenthem.Itisadvisable tocombinedirectexamsampleswithculturemethodsifknowingviabilityisimportanttoyourproject. • Tapeliftsarenotabletobecultured. • Ifadirectexaminationofaswabsampleistaken,afollowupbycultureispossible. • Directexaminationsofdirt/soilanddustsamplescannotbeperformedreliablybecauseofpreparationlimitations. • Fungiusuallycannotbeidentifiedtospeciesandsometimesnoteventogenuswiththismethod.Forexample, Aspergillus sp. and Penicillium sp. are normally reported together due to the similarities in spore morphology, unless fruiting structures are present that allows for a better identification.

Materials For Tape Lift: • WerecommendusingEMSLTapekitsorBio-Tapes(ZefonIntl)otherwiseclear (transparent) Scotch or other brand tape (frosted tape obscures the spores). • Newplasticbagtoholdsample(s)(providedinatapeliftkitavailablefromEMSL). • Onlyusetapeliftsonhard,flatsurfaces For Bulk: • Sterilecontainer,newplasticbagorWhirl-Pak(providedbyEMSL) to hold and transport samples. For Swab: • SterileTransPorterswabtocollectandtransportsamples(providedbyEMSL). For all Matrices: • Latex/nitrilegloves(alsocanbeprovidedatyourrequest).

Sampling Procedure

Tape-lift 1.Takeafewinchesofcleartape.Avoidtouchingthestickyside,especiallythe part to be used to collect the mold. 2.Wearinggloves,applythecentralinchoftapetothesuspectarea(chooseone thatisfreeofextraneousdebris).Applylightpressuretothenon-adhesiveside. 3. Pull tape off surface with slow, steady pressure, holding the tape edges only. 4.Applystickysideoftapetotheinsideoftheplasticbag(ziplock). 5. Ensure there are no folds or creases in the tape. 6. Close bag and label appropriately. (Put only one sample in each bag.)

Sampling Procedure 1. Makesurethehoseattachmentisconnectedtoyourvacuumcleanerproperly.Turnonthevacuumcleanerto makesurethehoseattachmentispullingair,andthenturnoffthevacuumcleaner.Sometimesthedust collectiondevicewillnotfitcorrectlyontothehoseattachment.Ifthisisthecase,youmayuseducttapeor electrical tape to tape the dust collector to the hose attachment. 2. Usetheextensioncordasneededtoreachthemarkedareawiththevacuumhose. 3. Removebothcapsfromsamplingdevice.Placethecapsinalocationsoyoucanfindthemafterthetestiscompleted. Attachtheflat,roundendofthesamplerdevicetotheendofthehoseattachmentofyourvacuumcleaner. 4. Usetheslantedendofthesamplingdevicetocollectyoursample.Keeptheslantendofthesamplingdeviceflush with the surface to be sampled. 5. Turnonthevacuumcleanerandstartthewatchortimer.Starttimingthevacuumingprocedureusingthestopwatch. Try not to disturb the tape. Do not exceed the 5 minute sampling period. 6. Vacuumtheareacontainedwithintheductorelectricaltape.Dothisbypassingthesamplingdeviceoverslightly overlapping,imaginaryparallellineswithinthesamplingareaforabout5minutes.Ifnecessary,adjustyourrateof movementsothatatotalof5minutesisusedtovacuumtheentire18squarefootsamplingarea. 7. Movetothesecondroomandrepeatthevacuumingofthetargetarea.Afterthesamplingiscompleted,holdthe samplingdeviceupwardtowardtheceilingandturnoffthevacuumcleaner.Re-captheslantendofthesampling devicesoasnottolosethedustcollected. 8. Avoidvacuumingupanylargedebristhatisnotdust.Ifyouaccidentallysuckupthetape,pointthesamplingdevice towardtheceilingandturnoffthevacuumcleaner.Pickthetapeoutofthesamplingdevice.Turnthevacuumcleaner backonandreturntovacuumingthesamplingarea.Besuretoaccountforlostsampletimewhenyoudothissoyou get a total of 5 minutes of sampling time. 9. Separatethesamplingdevicefromthehoseofthevacuumcleanerandre-captheflatendofthedevice. 10. Afterthesmallcapsaresecuredonthedustsamplingdevice,makesurethereisdustinthesamplingcontainerbefore yousendittothelab.Ifnovisibledustisnoticed,repeatthesamplingprocedureinbothroomsindifferentlocations untilvisibledustispresentinthedevice. 11. Water Sampling for Total Coliform, E. coli,&FecalColiform

Sample Collection • Allwatersamplesmustbetakeninapproved,sterilesamplingcontainers, whichourlaboratorycanprovideuponrequest.Treated(municipality/ distribution systems) water samples must be de-chlorinated with sodiumthio-sulfatewhichisprovidedinthe100mLlabissuedbottles. • Keepsamplingbottlecloseduntilitistobefilled.

• Methicillin-resistantStaphylococcus aureus, (MRSA) is a type of bacteria that is resistant to treatment with antibiotics related to penicillin. This bacterium is generally spread through direct contact with the hands of a health care worker or person who is infected or carrying the organism. Endotoxin Sampling

• Endotoxinsarecompoundsthatarefoundinthecellwallsofgramnegativebacteria.Endotoxinscancause flu-likesymptomswheninhaledandmayaggravateasthma. • Highlevelshavebeenreportedfromavarietyofenvironments,suchasinrecycledwater,cottonmills, agriculture or wastewater treatment facilities, industrial wastewater mists, and contaminated room humidifiers. HighlevelsofEndotoxinsarefoundinanyrecycledfluid.

Summary of Procedures • Airsamplesarecollectedonendotoxin-freemembranefilters,storeddesiccatedwhereverpossible,orstored at 4°C. Endotoxin analysis can be done on air, water, or bulk samples.

Air Sampling 1. Cautionmustbetakentoavoidbreathingon,touching,orotherwise exposing the sampling containers to human contamination. 2. For indoor sampling, ensure that all doors and windows are closed. 3. Calibrate pump to 1.5 liters per minute 4. Usingglove,connectcassettetopumpwiththered-cappedendtoward the pump. Collect air samples for 4 hours per sample. 5. Disconnectcassetteandreplacetheprotectivecoveringbackonboththe inlet and outlet of the cassette, and wrap entire cassette in original packing and seal with tape.
