mitochondria

appear as black dots or rods in the basal aspect (attached side) of these tubule cellsStained as dark black spots due to high acid content. Red arrowUnder .22um in width, cannot actually be resolved with Light Microscope1Mitochondria in Kidney:Black staining of the mitochondriaMore black staining means more mitochondriaBottom of kidney slide lots = more transportTop has little = lighter = less transportMitochondria have cristaeMitochondria are rod like and about 0.25 um in diameter (can be 7 um in length)

Mt on EM micrograph

Intermembrane space (Black Arrow)Cristae (Red Arrow)Matrix (Green Arrow)Mitochondrial DNA is difficult to make out

3Nissl bodies in neurons

Nucleus with darker purple staining nucleolus (site of rRNA synthesis)Purple staining is the ribosome attached to Rough Endoplasmic reticulumNucleolus stained darkly bc it is the site of r-rna synthesis

4Ribosomes and RER in spinal nervous tissueCytoplasm is filled with this blue staining material = RER and RibosomesThe stain actually is staining the ribosomal RNA in the ribosomes on the RERThe nucleolus is also highly stained because this is where ribosomes are constructed***Do not rely on color to make determinations because the parts being stained could be dispersed, concentrated, etc which will cause different coloringShould probably understand what each stain marks/ binds to

Golgi apparatus

Small horse-shoe shaped organelle around periphery of nucleusThese are usually in the region very near the nucleus. The nucleus is unstained and can be visualized as a round clear area surrounded by black flecks (Golgi) in the cytoplasm. Note the crescent shaped Golgi structure. This organization is much more clearly observed in the EM demonstration micrographs.

6Golgi apparatus in Dorsal Root Ganglion SlideThe Nucleus is a clear colorSurrounded by black dots which may look crescent shapedThese are the Golgi apparatusesGolgi are perinuclear (only around the nucleus)Can see very discretely unlike the ribosomesSpecific silver stain that only differentiates Golgi

G.A. on EM micrograph

8Microfilaments and Skeletal MuscleSome microfilaments stain dark and then adjacent to this there are other microfilaments that stain lighter which causes the appearance of bands

Direction of ContractionStriated tissue seen as light, vertical bands. Microfilaments actually run horizontallyA-Band, Seen as darker vertical band, composed of myosin. (Stationary during contraction)All considered inclusions. Not membrane bound and low metabolic activity

NucleiUse nuclei to identifiy cell typeUse size, location, and shape to determine cell type

Nucleus and the LiverCan see difference in the nucleus of the hepatocyte and the endothelial cell

Neurovascular bundle and nucleus (Nerve, artery, and vein run together)

Skeletal muscles and nucleiNuclui located on the periphery Can have many nucleiAlso see microfilaments (pink) in muscle cell

Blastula and Mitosis (Nucleus examination)Black dots are yolk in blastula ignore it

Unit 1 Exam Microanatomy Lab Practical Practice QuestionsCytology, Epithelium, and Connective Tissue

What Cellular Organelle can be visualized by this Regauds Stain for Iron (blue and Black arrows?

Answer: Mitochondria21

What is the function of the organelle stained within the red circle?Answer: Protein Synthesis (Ribosomes and Rough ER make up the Nissl Bodies in these Neurons)22What membrane bound organelle indicated by the blue arrows appears as these dark moon shaped structures in this type of staining and is involved in packaging of proteins into vesicles?

Answer: Golgi Aparatus23Which phases of the cell cycle are indicated by the colored numbers

12345Answer: 1 = Anaphase2 = Prophase3 = Metaphase4 = Prophase5 = Telophase/ cytokinesis24What is the function of this cellular organelle?

Answer: Energy Production25What is the organelle that is show in this Micrograph that is involved in the production of protein?

Answer: Ribosomes or Polysomes26The coated pits and coated vesicles in this micrograph are surrounded by what protein?

Answer: Clathrin27These cytoplasmic granules indicated by the green arrows are utilized by the mitochondria because they supply energy in the form of what?

Answer: Sugars (the inclusions are glycogen)28This double membraned organelle regulates signals to the entire cell what structures indicated by the red and green arrows allow them send out these signals?

Answer: Nuclear Pores29
