The N a t i o n a l I n s t i t u t e o f Diabe tes and D i g e s t i v e and Kidney Diseases ( N I D D K ) announces t h e e s t a b l i s h m e n t o f a Human L i v e r C e l l C u l t u r e F a c i l i t y a t SRI I n t e r n a t i o n a l , Menlo Park, C a l i f o r n i a , f o r t h e purpose o f making human h e p a t o c y t e s more widely a c c e s s i b l e t o r e s e a r c h e r s . The F a c i l i t y i s now prepared t o begin t h e f irst phase o f p r o v i d i n g h e p a t o c y t e s t o s e l e c t e d i n v e s t i g a t o r s .

N I D D K s u p p o r t s t h e a c q u i s i t i o n o f e x c e s s normal human l i v e r s from organt r a n s p l a n t donors and t h e i s o l a t i o n and c u l t u r e o f h e p a t o c y t e s from t h e s e t i s s u e s . Human l i v e r c e l l s i n h igh y i e l d are b e i n g r o u t i n e l y p r e p a r e d .Q u a l i t y c o n t r o l d a t a on i n i t i a l v i a b i l i t y , s u r v i v a l i n c u l t u r e , s e l e c t e d l i v e r s p e c i f i c f u n c t i o n s , and donor informat ion ( e . g . , age, sex, race, c a u s e o f d e a t h ) are a v a i l a b l e .

The human l i v e r c e l l s can b e used e i t h e r i n t h e F a c i l i t y by a v i s i t i n gi n v e s t i g a t o r who is r e s i d e n t a t t h e l a b o r a t o r y o r who comes o n l y when ce l l s are a v a i l a b l e . S e l e c t e d experiments w i t h human h e p a t o c y t e s can be conducted f o r r e s e a r c h e r s by F a c i l i t y personnel , when t i m e permi ts , b u t t h e c o s t s o f t h e s e s p e c i a l a s s a y s are n o t covered by t h e N I D D K suppor t and must be reimbursed. The pr imary s t r a i n o f mice t o be examined i s t h e B6C3F1 mouse though o t h e r heterozygous rodent s t r a i n s may a l s o be c o n s i d e r e d . The N a t i o n a l I n s t i t u t e o f Deafness and Other Communication D i s o r d e r s ( N I D C D ) ,N I H , h a s a requirement t o have a c o c h l e a r implant des igned f o r use i n c h i l d r e n and which could be removed and r e p l a c e d without i r r e v e r s i b l e trauma t o t h e cochlea and a s s o c i a t e d t i s s u e s . T h i s implant w i l l u t i l i z e e l e c t r o d e a r r a y s .

P a s t s t u d i e s suppor ted by N I D C D and i t s p r e d e c e s s o r , t h e N a t i o n a l I n s t i t u t e o f Neurologica l and Communicative D i s o r d e r s and S t r o k e , have suppor ted t h e development o f e s s e n t i a l l y a l l components i n implanted a u d i t o r y p r o s t h e s e s .These have i n c l u d e d t h e e v a l u a t i o n o f t h e s a f e t y o f c h r o n i c i m p l a n t a t i o n , and e l ec t r i ca l s t i m u l a t i o n o f t h e c o c h l e a . For t h e most p a r t t h e s e components were des igned f o r permanent i m p l a n t a t i o n i n a d u l t s . Recent ly t h e N I D C D , w i t h t h e t e c h n i c a l a s s i s t a n c e o f t h e Neural P r o s t h e s i s Program, h a s suppor teds t u d i e s o f problems t h a t are unique t o implants i n c h i l d r e n . S p e c i f i c a l l y ,t h e e f fec ts o f head growth and r e c u r r e n t o t i t i s media have been e v a l u a t e d i n young an imals .

The r e s u l t s of t h e s e s t u d i e s i n d i c a t e t h a t f i b r o u s t i s s u e e n c a p s u l a t i o n of l e a d wires connec t ing e l e c t r o d e a r r a y s i n t h e s c a l a tympani w i t h implanted r e c e i v e r - s t i m u l a t o r s i n t h e mastoid r e g i o n can c a u s e s i g n i f i c a n t problems. Also, h i s t o p a t h o l o g i c a l examinat ions o f c o c h l e a s a f t e r c h r o n i c i m p l a n t a t i o n of e l e c t r o d e s have r e v e a l e d f i b r o u s t i s s u e , and i n some c a s e s , new bone growthoccluding t h e s c a l a tympani . Although it appears t h a t p r a c t i c a l d e s i g n s which w i l l accommodate head growth w i l l be p o s s i b l e , t h e replacement o f t h e s e d e v i c e s , should t h e y f a i l o r when improved a u d i t o r y p r o s t h e s e s become a v a i l a b l e , w i l l n o t be t r i v i a l . T h i s i s e s p e c i a l l y important i n c h i l d r e n because of t h e l o n g a n t i c i p a t e d p e r i o d o f c o c h l e a r implant use over t h e i r l i f e spans .

The N a t i o n a l I n s t i t u t e o f Neurologica l D i s o r d e r s and S t r o k e (NINDS) h a s a requirement t o e v a l u a t e t h e s t r u c t u r e - a c t i v i t y r e l a t i o n s h i p s (SARI f o r a n t i c o n v u l s a n t drugs screened by t h e A n t i e p i l e p t i c Drug Development ( A D D )Program o f t h e NINDS.

The E p i l e p s y Branch, D i v i s i o n of Convulsive, Developmental and Neuromuscular D i s o r d e r s , NINDS, conducts an e x t e n s i v e A n t i e p i l e p t i c Drug Development ( A D D )Program aimed a t i d e n t i f y i n g p o t e n t i a l l y new a n t i e p i l e p t i c a g e n t s t o be used i n man. The ADD program i s an ex t ramura l program r u n through t h e c o n t r a c t mechanism which was e s t a b l i s h e d as a c o l l a b o r a t i v e e f f o r t w i t h academia, government, and t h e pharmaceut ica l i n d u s t r y i n o r d e r t o develop more e f f e c t i v e and l e s s t o x i c a n t i - c o n v u l s i v e t h e r a p e u t i c a g e n t s . T h i s program employs a s tep-wise approach u t i l i z i n g a systematic ser ies o f s c r e e n i n g tests and d e c i s i o n s t e p s t o advance c a n d i d a t e compounds through t h e s t a g e s o f p r e c l i n i c a l and c l i n i c a l development. Compounds t h a t demonstrate t h e r a p e u t i ci n d i c e s are advanced t o sub-chronic o r a l t o x i c i t y . The C o n t r a c t o r w i l l be r e s p o n s i b l e f o r e s t a b l i s h i n g a d a t a b a s e t o a n a l y z e s t r u c t u r e - a c t i v i t yr e l a t i o n s h i p s f o r s y s t e m a t i c p r e d i c t i o n s of i n c r e a s i n g a c t i v i t y and/ord e c r e a s i n g t o x i c i t y . T h i s requirement r e p r e s e n t s a new component o f t h e ADD program t o conduct a n a l y s i s o f approximately 14,000 s t r u c t u r e s which w i l l be o r have been screened f o r a n t i c o n v u l s a n t a c t i v i t y .

The a n a l y s i s of q u a n t i t a t i v e s t r u c t u r e - a c t i v i t y r e l a t i o n s h i p s (QSAR) is based on c a l c u l a t i n g t h e o r e t i c a l chemical s t r u c t u r a l a t t r i b u t e s and s t a t i s t i c a l l ycomparing t h e chemical and b i o l o g i c a l d a t a . QSAR methods t h a t re la te f u n c t i o n s o f chemical s t r u c t u r e s t o measures o f b i o l o g i c a l a c t i v i t y w i l l be used t o develop t h e mathematical e q u a t i o n s o r s t r u c t u r e - a c t i v i t yr e l a t i o n s h i p s . These e q u a t i o n s w i l l be used by t h e Government t o p r e d i c ta c t i v i t y o f new a g e n t s d i r e c t l y coming i n t o t h e ADD program. Algori thms w i l l be c o n s t r u c t e d around t h e mathematical e q u a t i o n s f o r u s e on a day-to-day b a s i s t o a i d i n t h e d e s i g n and s e l e c t i o n of drugs o f h i g h e r potency and lower t o x i c i t y . Purpose

The purpose o f t h i s c o o p e r a t i v e agreement i s t o suppor t a drug abuse t r e a t m e n t r e s e a r c h demonst ra t ion program i n t h e Dis t r ic t o f Columbia. T h i s program w i l l t e s t and e v a l u a t e t h e e f f e c t i v e n e s s of v a r i o u s drug abuse t r e a t m e n t approaches and combinat ions o f t r e a t m e n t methods and approaches i n o r d e r t o examine t h e i r d i f f e r e n t i a l e f f e c t i v e n e s s i n p a r t i c u l a r d i a g n o s t i c and p a t i e n t / p o p u l a t i o n subgroups. I t is a l s o expected t h a t funds a v a i l a b l e from t h i s p r o j e c t w i l l be used t o expand t r e a t m e n t c a p a c i t y and t h e number o f t r e a t m e n t s l o t s a v a i l a b l e i n t h e Distr ic t o f Columbia.

The Di s t r i c t o f Columbia was s e l e c t e d a s t h e s i t e f o r t h i s model t r e a t m e n t r e s e a r c h demonst ra t ion p r o j e c t because o f t h e e x t e n t and i n t e n s i t y o f t h e drug abuse problem, a v a i l a b i l i t y o f c l i e n t s , and because t h e combinat ion o f p a t i e n t subgroups and t y p e s o f drug abuse problems l e n d s i t s e l f t o a r e s e a r c h demonst ra t ion p r o j e c t t h a t may he a b l e t o b e r e p l i c a t e d i n o t h e r p a r t s o f t h e c o u n t r y . A s p a r t o f t h e N a t i o n a l Drug Cont ro l S t r a t e g y , r e l e a s e d i n September1989, Washington, D . C . was i d e n t i f i e d as t h e f i r s t s i t e f o r a model drug t r e a t m e n t demonst ra t ion program.

A p p l i c a t i o n s are be ing s o l i c i t e d i n t h i s Request f o r A p p l i c a t i o n s ( R F A ) f o r two components: ( 1 ) a Diagnos t ic , R e f e r r a l , and Data Management Unit and ( 2 ) two o u t p a t i e n t t r e a t m e n t programs, each o f f e r i n g a d i f f e r e n t range o f s e r v i c e s . 11. MECHANISMS OF SUPPORT

Support w i l l b e provided through c o m p e t i t i v e l y awarded c o o p e r a t i v e agreements .N I A I D h a s set a s i d e $3 ,632 ,000 i n t o t a l c o s t s f o r t h e i n i t i a l y e a r ’ s funding t o m i n o r i t y i n s t i t u t i o n s . The N I A I D p l a n s t o fund between 4 and 5 awards from t h i s announcement. The s t a r t i n g d a t e f o r t h e i n i t i a l annual p e r i o d w i l l be on o r b e f o r e September 1990. Awards w i l l be made f o r a p e r i o d o f 3 y e a r s w i t h y e a r l y e v a l u a t i o n i n v o l v i n g competent c o n s u l t a n t s .
