Enabling Technologies for

Cancer Research

Analysis of tumor suppressor gene methylation with

the LightCycler® 480 System. Mixtures of unmethylated

and fully methylated genomic DNA were treated with

bisulfite. A fragment of the MGMT tumor suppressor gene

was then amplified and analyzed by high-resolution

melting. This allowed semi-quantitative determination of the

portion of methylated DNA contained in each case.

Biological Sample ConcentrationThe miVac Quattro is a versatile, high capacity biological sample concentrator that rapidlyremoves a wide range of solvents, from volatile organic solvents to water. It is suitable foruse in applications including oligonucleotide synthesis, RNA and DNA preparation, peptidepreparation, sequencing, molecular biology, and absorption, distribution, metabolism,excretion, toxicity (ADMET) research. A high displacement pump removes water quickly andquietly. Solvent removal can be enhanced by the addition of a miVac SpeedTrap refrigeratedcondenser. The four-swing position rotor can simultaneously accommodate up to 20

shallow-well microplates or eight deep-well plates. A wide variety of rotors is available toaccommodate formats including tubes, microplates, and vials. The instrument offers digitalcontrol of concentrator temperature and short concentration times, reducing the risk of heatdamage to samples.

Instrument-Free DNA DetectionThe BioHelix Express Strip Cassette is a novel, disposable DNAdetection device. It is designed for instrument-free, cross-contamination-proof detection of amplicons derived frompolymerasechain reaction, helicase-dependent amplification, and other nucleicamplification reactions. The amplicons are dual-labeled by biotinand fluorescein isothiocyanate during amplification and detected ona test strip embedded in the device. Protein-DNA Binding AssayThe Protein-DNA Binding Assay provides a safe, fast, and sensitivealternative to traditional electromobility shift assays for detection andquantitative characterization of protein-DNA interactions. This assayeliminates the need for radioactive labeling and gel electrophoresisbecause it features chemiluminescent ProLabel detection and canbe performed in a 96-well plate. A small (about 6 kDa) ProLabeltag is fused to the protein of interest to generate a fusion proteinthat is capable of producing a strong chemiluminescent signal viaan enzyme complementation assay.
