NEW PRODUCTS

Erivedge (vismodegib) is a lowmolecular weight, orally availableinhibitor o the Hedgehog pathway. TheHedgehog pathway signalling throughthe smoothened transmembrane protein(SMO) leads to the activation and nuclearlocalisation o GLI transcription actorsand induction o Hedgehog target genes.Vismodegib binds to and inhibits SMO,thereby preventing Hedgehog signaltransduction. Erivedge is indicated or thetreatment o adult patients with metastaticbasal cell carcinoma, or with locallyadvanced basal cell carcinoma wheresurgery and/or radiation therapy are notappropriate. Erivedge is contraindicatedin pregnancy; women o childbearingpotential, unless two reliable methodso contraception are being used duringtreatment and or 7 months ater the lastdose. It is also contraindicated in nursingmothers during the course o treatment andor 7 months ater the last dose because othe potential to cause serious developmentdeects in breasted inants and children.Erivedge is available as a 150 mg tablet in

packs o 28s.

GyMiso (misoprostol) is a syntheticanalogue o prostaglandin E1. In the evento an early termination o pregnancy,the combination o GyMiso used in asequential regimen ater miepristone

leads to an increase in the successrate and accelerates the expulsion othe conceptus. GyMiso is indicatedin emales o childbearing age or themedical termination o a developingintrauterine pregnancy in sequentialcombination with a miepristone 200mg tablet, up to 49 days o gestation.GyMiso is contraindicated in the ollowing

situations: i there is a contraindicationto medical abortion including: known orsuspected hypocoagulation diseases,treatment with anticoagulants; uncertaintyabout pregnancy age; suspected ectopicpregnancy and contraindication tomiepristone. GyMiso is available as a 200microgram tablet in packs o 4s.

Jakavi (ruxolitinib phosphate) is aninhibitor o the Janus Associated Kinases(JAKs) JAK1 and JAK2 with nanomolarpotency. JAKs mediate the signalling o anumber o cytokines and growth actorsthat are important or haematopoiesisand immune unction. Ruxolitinib inhibitsJAK-STAT signalling and cell prolierationo cytokine dependent cellular modelso haematological malignancies. Jakaviis indicated or the treatment o diseaserelated splenomegaly or symptoms inpatients with primary myelobrosis, post-polycythemia vera myelobrosis or post-essential thrombocythemia myelobrosis.Jakavi is available as 5 mg, 15 mg and 20mg tablets in packs o 56s.

Priligy (dapoxetine hydrochloride)is a SSRI. Priligy is indicated or thetreatment o premature ejaculation (PE)in men 18 to 64 years o age, who haveall o the ollowing: an intravaginalejaculatory latency time (IELT) o less thantwo minutes; a persistent or recurrentejaculation with minimal sexual stimulationbeore, on, or shortly ater penetration

and beore the patient wishes; a markedpersonal distress or interpersonaldiculty as a consequence o PE; anda poor control over ejaculation. Priligyis contraindicated in all o the ollowingconditions: signicant pathological cardiacconditions (eg NYHA class II-IV heartailure), conduction abnormality (2nd, 3rddegree A-V block or sick sinus syndrome)not treated with permanent pacemaker,signicant ischaemic heart, valvulardisease; concomitant MAOIs, thioridazine,SSRIs, SNRIs, TCAs, serotonergicdrugs/ herbs (eg L-tryptophan, triptans,tramadol, linezolid, lithium, St Johnswort) (including less than 14 days beorestarting, less than 7 days ater ceasingdapoxetine); concomitant potent CYP3A4

inhibitors eg ketoconazole, itraconazole,atazanavir, nelnavir, ritonavir, saquinavir,telithromycin, neazodone; and in moderateto severe hepatic impairment. Priligy isavailable as a 30 mg tablet is packs o 3sand 6s.

Zaltrap (aibercept (rch)) is arecombinant protein consisting osequences derived rom human vascularendothelial growth actor (VEGF) receptor(1 and 2) extracellular domains used tothe Fc portion o human immunoglobulinG1 (IgG1). Afibercept is produced byrecombinant DNA technology in a ChineseHamster Ovary (CHO) K-1 mammalian

expression system. Zaltrap in combinationwith irinotecan-fuoropyrimidine basedchemotherapy is indicated in adults withmetastatic colorectal cancer previouslytreated with an oxaliplatin containingregimen. Zaltrap is contraindicatedor ophthalmic/ intravitreal use due tohyperosmotic properties o Zaltrap. Zaltrapis available in vials o 100 mg/4 mL and200 mg/8 mg.

Zarzio (flgrastim (rbe)) is arecombinant Human Granulocyte ColonyStimulating Factor (rmetHuG-CSF) derivedrom E. coli. Zarzio is indicated in thereduction o neutropenia duration, clinicalsequelae ater induction/ consolidationchemotherapy in AML; ollowingautologous or allergenic bone marrowtransplant. It is also indicated in themobilisation o peripheral blood progenitorcells (PBPC) ollowing myelosuppressivechemotherapy (autologous) or in normaldonors (allergenic); inection incidencereduction during myelosuppressive

chemotherapy in nonmyeloid malignancy;severe chronic neutropenia (SCN); and inthe maintenance o adequate neutrophilsduring antiretroviral/ myelosuppressiveHIV therapy. Zarzio is contraindicated inpatients with known hypersensitivity to Ecoli derived products. Zarzio is availableas a 300 microgram per 0.5 mL and 480microgram per 0.5 mL prelled syringe.

SAFETY RELATED CHANGES

Avapro, Karvea (irbesartan) andAvapro HCT, Karvezide (irbesartan/hydrochlorothiazide)are nowcontraindicated with aliskiren containingmedicines in patients with diabetes or withmoderate to severe renal impairment.

Eudix (uorouracil) is nowcontraindicated in women who are or maybecome pregnant during therapy.

Normison (temazepam) is nowcontraindicated and should not be usedas monotherapy to treat depression, orsymptoms o anxiety associated withdepression, due to a risk o suicide.

Xarelto (rivaroxaban) is nowcontraindicated in patients undergoingdialysis.
