Numerical aperture (NA)

Coverslip/Glass thickness

Magnifi cation

Tube length

Achromatic

Achromatic objectives are used in most microscopes. They bring light of different wavelengths to a focus producing a clearer image with less distortion and reducing chromatic aberration.

Body Tube Length

Distance between the objectives and the top of the body tube (usually 160mm). Objectives are designed to fi t a specifi c microscope. Incorrect objectives will result in aberration.

Coaxial Focus

A focusing arrangement where the fi ne and coarse focus knobs are situated together and share a common axis.

Condenser

The condenser focuses light upon a specimen under investigation. A built-in condenser has a fi xed focal length and a numerical aperture of 6.65. The Abbe condenser has a spiral focusing system and a numerical aperture of 1.25. To focus the condenser place a sharp object such a needle or a pencil onto the light source and adjust the condenser height until the object is in sharp focus. The numerical aperture (NA) of the condenser should be equal to, or greater than, the highest objective NA - usually 1.25 (for a 100x objective).

Compound Microscope

The “Classic Microscope”. Used to observe specimens and microscope slides in 2D. Using either transmitted or refl ected light, quite high magnifi cations may be obtained.

Chromatic Aberration

When a lens fails to bring light of differing wavelengths to a common focus colour fringes will be apparent around the edges of the image. This is known as Chromatic Aberration and can be reduced by using Achromatic Objectives.

Disc Diaphragm

A rotating disc situated beneath the stage. The disc contains fi ve or six apertures, of different sizes which are selected to control light levels and thereby contrast of the specimen. See also Iris Diaphragm.

Din

Abbreviation for Deutsche Industrie Normen. An industry standard for optics indicating a high quality lens.

Eyepiece

The primary image is formed by the objective lens - the eyepiece then forms a secondary image. There are two common eyepieces which differ in lens arrangement. The Huyghenain eyepiece has a similar fi eld of view than the Widefi eld or Ramsden.

All Philip Harris microscopes have locked in eyepieces for security.

Eyepiece Diaphragm

The diaphragm defi nes the round fi eld-of-view seen when looking down the microscope. It accepts graticules and is positioned at a point that ensures the graticule scale is in focus with the specimen image.

Field of View

Area that is visible through the eyepiece when the microscope is in focus.

Graticule

A glass or plastic disc with printed on scale. Graticules are mounted in the eyepiece, calibrated using a stage micrometer and allow accurate measurement of subjects under observation.

Immersion Objective

When high magnifi cation and high resolution are required it is necessary to exclude air, which can cause refl ection, between the microscope slide and the lower surface of the objective. Exclusion of air is achieved by using a drop of immersion oil which forms a light-tight connection between the two surfaces. A x100 oil immersion is necessary for viewing small specimens such as bacteria and cell organelles and must be used with an Abbe condenser.

Iris Diaphragm

A fully adjustable leaf diaphragm used to control the amount of light converging on the specimen, thereby altering contrast. See also disc diaphragm.

LED Illumination

A modern type found on an increasing number of microscopes. Ideally suited to education as it provides a very even illumination with excellent contrast. Cooler than Halogen, it is ideal for live specimens. Most LED units are rechargeable so there is no need for mains power during use.

Magnifi cation

The factor by which the image is larger than the object. The total magnifi cation is calculated by multiplying the magnifi cation of the objective by that of the eyepiece. See also resolution.

Plano-concave Mirror

Usually two-sided (50mm diameter), with one side fl at (plano) and the other curved (concave). Always use the plane surface with a microscope lamp. Use the concave surface for very low NA objectives and with natural light.

Objective Lens

These form the primary image of the object which is further magnifi ed by the eyepiece. The objective lens body will be marked as shown above.

Parcentred

The optical system (condenser, eyepiece and objective lens), are directly aligned to minimise aberration.

Parfocality (parfocal)

The property of a microscope which ensures they stay in focus when objectives are changed.

Polariser

An addition to a microscope which reduces light to a single plane revealing spectrum colours. Ideal for use with rock sections or other crystalline specimens.

Resolution

The resolving power of a microscope indicates the minimum distance between two points that can be distinguished. High magnifi cation does not mean high resolution. See also numerical aperture.

Retractable Objective

Spring-loaded objective that prevents damage to microscope slides should the objective be lowered too far.

Slip Clutch

A safety mechanism to prevent gear stripping if the coarse focus is racked down beyond the normal stop position.

Stage

A platform upon which the microscope slide is placed. The slide is usually held in place by stage clips in which case the slide can be moved around manually. For more accurate positioning of slides a mechanical stage can be used.

Stereo Microscope

Used to observe specimens and larger objects in 3D, such as fungi, fossils, crystals. Also used in microdissection. The magnifi cation range is lower than a compound microscope, typically x20 to x80.

Numerical Aperture (NA)

The Numerical Aperture of the lens determines its ability to resolve fi ne detail. The greater the NA value of the optical system (condenser and objective lens), the greater the resolution. (Numerical apertures are marked on both the condenser and the objective.)

Monocular

1. What do you want to observe?

2D Images e.g. microscope slides:

Compound Microscope.

• View slide-mounted specimens that are thin enough to allow light to pass through them

• Examine premounted slides and pond-water specimens such as algae and protozoans

• Provide high-power magnifi cation (40x to 1,000x), a narrow fi eld of view and a short working distance

3D Objects e.g. fossils, microdissection:

Stereo Microscope.

• Observe unmounted, macro-sized specimens that are too thick to allow light to pass through them

• View insects, plants, rocks, coins and more

• Provide low-power magnifi cation (10x to 40x), a wide fi eld of view and a long working distance

2. Who will be using it?

Pre-16 Students:

For absolute beginners we recommend the Economy Microscope, however all of our Philip Harris Microscopes are designed to meet the needs of secondary school students.

Post-16 Students:

The Philip Harris Stereo Microscopes or any of the Compound Microscope range.

College/University Students:

Digital Compound Stereo Microscope or the Optika Monocular Ecovision Microscope.

Teacher/Demonstrator:

We recommend our Trinocular Microscope or any of our microscopes with integrated digital cameras.

3. What type of illumination do you need?

Low Magnifi cations (up to 100x):

Mirror or Built-in Light Source.

Higher Magnifi cations:

Mirror and Lamp or Built-in Light Source.

There are 2 types of built-in illumination available:

Tungsten:The most common and least expensive. Brighter than fl uorescent.

LED:Cool even light. Ideal for live specimens. Long life from 5000 - 10000 hours.

4. How will the microscope be used?

• For short periods of time

• For standard viewing

We recommend Monocular Head.

• For extended periods of time

• For comfort delivering long term viewing

We recommend Binocular Head (offers greater comfort and less eye strain).

• While user is seated:We recommend Inclined Head.

• While user is standing:We recommend Vertical Head.

• With a computer or television:We recommend Digital Microscope (please check specifi cations to ensure compatibility).

• With third person viewing (teacher or camera): We recommend Trinocular Head.
