1. Introduction

The ReliaPrep™ Large Volume Heater Shaker Magnet Instrument (ReliaPrep™ LV 32 HSM) is designedto perform all of the functions necessary for the processing of magnetic resin-based purifications incommon 50ml conical tubes. With its ability to heat, shake and apply a magnetic field, the ReliaPrep™LV 32 HSM Instrument provides all-in-one processing capabilities for a variety of large volumepurification chemistries in either a manual or automated format. The instrument uses standard 50mlconical tubes, magnets and reagent-based paramagnetic particles (PMPs). The PMPs provide a mobilesolid phase that optimizes capture, washing and elution of biological target molecules.

Initially designed to run the Promega ReliaPrep™ Large Volume HT gDNA Isolation System (Cat.# A1751), the ReliaPrep™ LV 32 HSM Instrument is supplied with preprogrammed isolationmethods for processing up to 32 samples of human whole blood in approximately 2–3.5 hours,depending on sample type and number. Samples are processed in a semi-automated method with theuser dispensing and aspirating reagents from the samples as directed by the LCD screen. Theprogrammed methods instruct the instrument when to apply shaking, heating and magnetic forces tothe sample tubes for cell lysis, capture and elution of DNA. The ReliaPrep™ LV 32 HSM Instrumentscan be controlled simultaneously on a robotic liquid handling workstation to fully automate processingof blood samples. The number of samples per instrument run depends on the number of Heater ShakerMagnets (HSMs) integrated. Each HSM is able to accommodate 1-32 samples. In both manual andautomated processing, high-quality and high-yield DNA is obtained for direct use in a variety ofdownstream analytical applications.

1.A. Intended Use of the ReliaPrep™ LV 32 HSM Instrument

The ReliaPrep™ LV 32 HSM Instrument is designed for use in combination with the ReliaPrep™ LargeVolume HT gDNA Isolation System for the isolation of genomic DNA from 3, 6, or 10ml of humanwhole blood. When controlled by a robotic liquid handling workstation, the ReliaPrep™ DNA isolationchemistry is scalable for samples from 3–10ml. While designed to work with the ReliaPrep™ LargeVolume HT gDNA Isolation System, the ReliaPrep™ LV 32 HSM Instrument has applicability to avariety of large volume paramagnetic resin-based purification chemistries. Use of this instrument withchemistries other than the ReliaPrep™ Large Volume HT gDNA Isolation System may requiredevelopment and optimization of manual or automated methods by the user.

The ReliaPrep™ LV 32 HSM Instrument is intended for indoor use only. If liquid gets inside theinstrument, it may damage the electronics. Install in a location that meets the following criteria:

• Install on a sturdy, dust-free, level surface.• Choose a location that has good air circulation and is not exposed to direct sunlight.• Avoid noisy electrical power sources (e.g., power generators).• Do not install in a location where there is large temperature variability or high humidity.• Position the instrument so that it is easy to unplug from the power source.•Do not place the power supply in areas where it may come in contact with liquid.• Do not place next to heat sources.• Do not use near flammable gases or liquids.• For optimal performance, we recommend equilibrating the instrument to room temperature before

operating.

3. Unpacking and Setting Up the ReliaPrep™ LV 32 HSM Instrument

3.A. Setup

1. Remove the ReliaPrep™ LV 32 HSM Instrument, Power Supply and accessories from theshipping containers.

Important: Save the packaging material (including the shipping pallet) in case the equipmentneeds to be returned for service or repair at a later date.

2. Check that all parts have been included. Refer to Sections 1.C and 1.D for a list of parts.

3. Set the ReliaPrep™ LV 32 HSM Instrument on a flat, level surface in a dust-free location withreasonable air circulation.

4. Ensure that the power switches are in the off position. The power switches are located on thefront of the ReliaPrep™ LV 32 HSM Instrument and the back of the Power Supply (Figures 3and 5, respectively).

5. Connect the ReliaPrep™ LV 32 HSM Instrument to the Power Supply using the blue PowerCable by plugging the cable into the back of the ReliaPrep™ LV 32 HSM Instrument (Figure4) and the back of the Power Supply (Figure 5).Important: Place the power supply in a dry location away from possible liquid spills.

6. Plug the black Power Cord into the back of the Power Supply (Figure 5). Plug the Power Cordinto a grounded wall outlet. See Section 1.B for power requirements.

7. Turn on the power switch located on the rear of the Power Supply (Figure 5). Turn on thepower switch on the front of the ReliaPrep™ LV 32 HSM Instrument (Figure 3).

8. The instrument will now display the firmware version number and operational setting andwill perform a self-diagnostic check.

9. Once all diagnostic checks have been passed, the ReliaPrep™ LV 32 HSM Instrument is readyfor operation.Notes:While the instrument can be controlled by a computer, an external computer is not requiredfor normal operation of the instrument. If the instrument fails any of the diagnostic checks, an error message will be displayed. Referto Section 4.F of this manual for more information or contact Promega Technical Services oryour authorized service representative for assistance.

3.B. Shutdown

Turn off the power switch on the front of the instrument, then turn off the switch on the back of thepower supply. Unplug the Power Supply from the electrical socket. Disconnect the ReliaPrep™ LV 32HSM Instrument from the Power Supply.

4. Operating the ReliaPrep™ LV 32 HSM Instrument

4.A. Main Menu Navigation

After successful self-diagnostic check, the Main Menu is displayed.

Menu Options

1. Remote. This menu option allows the instrument to be controlled by an external computerconnected via RS-232 cable to the port on the back of the ReliaPrep™ LV 32 HSM Instrument.

2. ReliaPrep. This menu option allows semi-automated processing of blood samples using theReliaPrep™ Large Volume HT gDNA Isolation System (Cat.# A1751).

3. Manual. This menu option allows manual control of the ReliaPrep™ LV 32 HSM Instrumentfunctions without connecting it to an external computer.

4. Setup. This menu option offers diagnostic testing and demo modes. See Section 7.A for details.

Use the "Up" and "Down" buttons to move the cursor to the desired menu choice. Once the cursor ismoved to the desired position in the list, press the "ENTER" button to make the selection. At any timeduring the selection process, you may press the "ESC" button to return to the previous menu.

4.B. Sample Purification Using the ReliaPrep™ Large Volume HT gDNA Isolation System

The ReliaPrep™ LV 32 HSM Instrument is designed to isolate genomic DNA from human whole bloodsamples using the ReliaPrep™ Large Volume HT gDNA Isolation System. Please refer to the ReliaPrep™Large Volume HT gDNA Isolation System Technical Manual (#TM341) for detailed instructions regardinginstrument sample handling, setup and genomic DNA isolation. Before starting a run, please ensure thatyou have sufficient reagents available for the entire run.

The ReliaPrep™ LV 32 HSM Instrument is designed for use with potentially infectious materials. Usersshould use the appropriate protection (i.e., gloves, goggles, etc.) when handling potentially infectionssubstances. Users should adhere to their institutional guidelines for the handling and disposal of allinfectious materials. If you are processing less than 32 samples, we recommend placing empty 50mltubes in unused positions to reduce the chance of instrument contamination.

The ReliaPrep™ Large Volume HT gDNA Isolation System reagents contain potentially hazardouschemicals. Users should wear protective gloves or use other protective means when handling thereagents. Users should follow their institutional guidelines for disposal.

Starting a Purification Run

1. After the instrument has been set up and the diagnostic tests completed (Section 3.A) followthe instructions in Section 3.A to set up the instrument prior to the DNA isolation run.

2. Using the scroll “down” button, select option 2 “ReliaPrep” from the Main Menu.

3. Using the scroll buttons, choose the appropriate protocol (3, 6 or 10ml blood).

4. Follow the prompts on the screen to process samples or refer to the ReliaPrep™ Large VolumeHT gDNA Isolation System Technical Manual #TM341.

4.C. Remote Menu Option

The Remote menu option allows the ReliaPrep™ LV 32 HSM Instrument to be controlled by an externalcomputer through the RS-232 port. For fully automated sample processing, the ReliaPrep™ LV 32 HSMInstruments may be integrated with a robotic liquid handling workstation. In this configuration theinstruments are all connected to a computer and controlled by a driver in the workstation software. Fordetailed information on integrating the ReliaPrep™ LV 32 HSM Instrument with a liquid handlingworkstation, please contact [email protected].

4.D. ReliaPrep™ Menu Option

The ReliaPrep™ LV 32 HSM firmware contains preprogrammed methods that can be used to process 3,6 or 10ml of blood. The instrument provides processing instructions through the LCD screen. Pleaserefer to the ReliaPrep™ Large Volume HT gDNA Isolation System Technical Manual (#TM341) for detailedinstructions.

4.E. Manual Menu Option

The ReliaPrep™ 32 LV HSM Instrument can be operated manually, such that shaker (speed, directionand duration), temperature and magnet (on/off) are controlled independently.

This instrument is designed for use with ReliaPrep™ nucleic acid isolation reagents. Use with any otherreagents must be developed, optimized and validated by the user.

When operating the ReliaPrep™ 32 LV HSM in manual mode, care must be taken to avoid liquid spillsand aerosols. When testing a new liquid or shaking speed, the optimal RPM setting should bedetermined by shaking at a minimum speed and increasing until the desired mixing is obtained. Table 1provides the maximum shaking speeds determined for two solutions, water and 50% ethanol.

Diagnostic Test

The “Diagnostic Test” option verifies that the heater, shaker and magnets are performing within thespecifications for the instrument. The message "Diagnostic Testing Please Wait…" is displayed on theLCD screen. The Setup Menu will be displayed after the diagnostic testing is completed and no errors areidentified. An error message will be displayed if the diagnostic testing identifies a parameter outside ofthe acceptable range for the ReliaPrep™ LV 32 HSM Instrument. If this happens, one of the followingerrors will be displayed: “Shaker/Sensor Error”, “Magnet/Sensor Error” or “Heater/Sensor Error”. Turnoff the instrument, wait for 10 minutes and turn the instrument back on. If the error message continues tobe displayed then contact Promega Technical Services for additional assistance.

Reliability Test

The “Reliability Test” option is used only during manufacture and testing of the ReliaPrep™ LV 32 HSMInstrument and is not intended for general use.

ReliaPrep™ Demo

The ReliaPrep™ Demo includes all the steps of the ReliaPrep™ HT gDNA Isolation System, but theduration of steps is reduced for demonstration purposes, making it unsuitable for DNA isolation. Thisprogram is for demonstration purposes only and should not be used for DNA isolation.

5. ReliaPrep™ Instrument Cleaning and Maintenance

The ReliaPrep™ LV 32 HSM Instrument requires minimal maintenance. However, it is important toclean the instrument at regular intervals. Spilled samples or reagents should be cleaned up immediately.Most parts of the instrument have an anodized coating, which forms a durable, easily cleaned barrier.However, if liquid gets inside the instrument, it may damage the electronics.

Always turn off and unplug the ReliaPrep™ Instrument or the Power Supply before cleaning.

No additional maintenance is required for the ReliaPrep™ Instrument, Power Supply or accessories.The Instrument and Power Supply do not contain any user-serviceable parts. Removing the Instrumentor Power Supply casing will void the warranty.

General Care

Wipe up any spills immediately.

After each use, clean the instrument by wiping off the top and sides of the ReliaPrep™ Instrument andthe Tube Rack using a cloth dampened with deionized water or 70% ethanol. Do not use other solventsor abrasive cleaners because they can damage the instrument’s anodized coating.Note: Wear gloves. If the instrument is used with biohazardous materials, dispose of any cleaningmaterials used in accordance with your institutional guidelines.

Keep the cooling vents in the back of the machine clear of dust.

If the inside of the ReliaPrep™ Instrument becomes contaminated with samples or reagents, remove theTube Rack and Cover to gain access to the tube holders and the magnet bar. The cover can be removedby loosening the eight screws that attach the cover to the instrument (Figure 6). Once the cover is

removed, wipe the tube holders and the magnet bar using a cloth dampened with deionized water.Wipe off the magnet bar and the tube holders using a cloth and foam- or cotton-tipped swabsdampened with 70% ethanol followed by a cloth and foam- or cotton-tipped swabs dampened with a1–2% bleach solution in deionized water. Follow immediately with a cloth and foam- or cotton-tippedswabs dampened with deionized water to remove any residual bleach from the instrument surfaces.Repeat the procedure as many times as required to effectively disinfect and decontaminate theinstrument.

• Keep the cooling vents in the back of the instrument and top of the Power Supply clear of dust.

7. Appendix

7.A. Instrument Startup, Diagnostics and Errors

When instrument is powered on:

Each time the instrument is turned on, it will automatically perform a self-diagnostic test during whichthe platform, heating elements, and magnetic assemblies are initialized to check that the instrument isproperly calibrated. If the diagnostic testing is successful, the LCD screen will display, “DiagnosticCheck Successful”, and after a short pause the instrument will transition to the main menu screen.

An error message will display if the diagnostic testing identifies a parameter outside of the acceptablerange for the ReliaPrep™ LV 32 HSM Instrument. If a “Calibration Error” is detected, turn off theinstrument, wait for 10 minutes and turn the instrument back on. If the error persists, contact Promegatechnical services for additional assistance.

During operation, the instrument will automatically do periodic self-diagnostic tests to verify that theplatform, heating elements and magnetic assemblies are within calibration. If the instrument detects thatany of these parts are not within calibration during a run, the method will pause and display“Calibration Error”. If a “Calibration Error” occurs during run, contact Promega Technical Services.

7.B. Updating Firmware

As new purification kits and applications become available, new versions of firmware may be required.The firmware version installed on your instrument can be verified either by accessing the FirmwareVersion option from the Setup Menu (see Section 4.F) or by turning the machine off and then on again.

Firmware is updated using the Reliaprep.exe program that will be supplied with any firmware updatesand the RS-232 port on the back of the instrument. Alternatively, the instrument can be connected to aUSB port on the computer using the included USB-to-serial converter cable. Firmware can be updatedfrom any computer running Microsoft Windows® 95 or higher with an available serial port or by usingthe included USB-to-Serial converter cable.

1. Plug the provided RS-232 Cable into the back of the instrument and into a serial port on yourcomputer. Note the address of the COM port that is being used.

2. Obtain the firmware upgrade program from Promega or your local Promega representative.Save the file to your hard drive. Within the Reliaprep.exe software, first select the Com portto which the shaker is connected, and then select “connect”. Select “Upgrade firmware” tostart the firmware upgrade process. As part of this process you will browse to the savedlocation of the firmware upgrade that you have downloaded. Once you have selected thefirmware upgrade program to be used for the upgrade, select “upgrade”. Upgrading thefirmware may take a few minutes. After completing the upgrade process, a message will bedisplayed indicating that the firmware has been upgraded successfully. At this point you canquit the Reliaprep.exe program.

Please contact Promega or your local Promega representative for assistance if you encounter anyproblems during the firmware upgrade process.

7.D. Returning the ReliaPrep™ LV 32 HSM Instrument and Accessories

The ReliaPrep™ LV 32 HSM Instrument is designed to provide years of consistent performance with littlemaintenance. If a problem arises with your instrument, please contact Promega or your local Promegarepresentative for support. If further action is required, repair options will be presented and a returnauthorization assigned if necessary. Promega is not responsible for instrumentation returned without anauthorization number. When you ship the instrument for service, please remember to:

1. Obtain a return authorization from Promega.

2. Decontaminate the instrument (see Section 7.E for Decontamination Instructions).

3. Affix a signed and dated Certificate of Decontamination to the outside of the package in which theinstrument is returned (see Section 7.E). Failure to complete and attach the Certificate ofDecontamination will result in a decontamination charge.

4. Use the original packaging to ensure that no damage will occur to the equipment during shipping.Any damage will incur additional charges. Notes:If any of the original packaging is lost or damaged, contact Promega or your local Promegarepresentative for replacement packaging.The original packaging includes the shipment pallet that the instrument was shipped on originally.Please use the same pallet for the return shipment.

5. Repack the equipment according to the following instructions.

Preparing the ReliaPrep™ LV 32 HSM Instrument and Power Supply prior to repacking:

1. Ensure that the tubes are removed from the instrument platform.

2. Ensure that the Power Supply is turned off and not plugged in.

3. Ensure that the ReliaPrep™ Instrument is turned off.

4. Make sure that the instrument is disconnected from the Power Supply.

Repacking the instrument:

1. Place the instrument back into the plastic bag.

2. Place the protective cardboard pieces over the ReliaPrep™ LV 32 HSMInstrument and over the Power Supply.

3. Repack the ReliaPrep™ LV 32 HSM Instrument accessories.

a. Rewrap the Tube Rack, Tube Rack Stand and Power Supply in bubblewrap and place in the appropriate location in the shipping container.

b. Place the RS-232 Cable, the Power Cable and the Power Cord intoresealable bags, and place the bags into the accessories box. Place the accessories box into the shipping container.

6. Affix the Certificate of Decontamination on the outside of the shipping box. Write the returnauthorization number provided to you by Promega or your local Promega representative on theoutside of the shipping box. Seal the box securely and send back on the original pallet provided.

7.E. Certificate of Decontamination

Disinfection and decontamination are required prior to shipping the instrument and instrumentaccessories for repair. Instruments returned must be accompanied by a signed and dated Certificate ofDecontamination attached to the outside packaging of the instrument.

To disinfect and decontaminate: Wipe off the outside surfaces using a cloth dampened with 70% ethanolthen a cloth dampened with a 1–2% bleach solution in deionized water. Follow immediately with a clothdampened with deionized water to remove any residual bleach from the instrument surfaces. Removethe Tube Rack (Figure 1) and the cover (Figure 5) to expose the Magnet Bar and the Tube Holders. Wipeoff the Magnet Bar and the Tube Holders using a cloth and foam- or cotton-tipped swabs dampenedwith 70% ethanol followed by a cloth and foam- or cotton-tipped swabs dampened with a 1–2% bleachsolution in deionized water. Follow immediately with a cloth and foam- or cotton-tipped swabsdampened with deionized water to remove any residual bleach from the instrument surfaces. Repeat theprocedure as many times as required to effectively disinfect and decontaminate the instrument.

Failure to confirm disinfection and decontamination will result in decontamination charges before theinstrument will be serviced.Select either (A) or (B):

A. I confirm that the returned items have not been contaminated by body fluids or by toxic,carcinogenic, radioactive, or other hazardous materials.

7.F. Warranty Information

Limited Warranty and Service Guidelines (Pending Warranty and Service Contract Information)

Promega warrants to the original purchaser that the Promega ReliaPrep™ LV 32 HSM Instrument willbe free from defects in materials and workmanship for a period of one year from the date of delivery.Promega agrees, as its sole responsibility under this limited warranty and upon prompt notice of adefect, to repair or replace (at Promega’s discretion) any instrument discovered to be defective withinthe warranty period. Expendable items are not covered by this warranty. This warranty does not includerepair or replacement necessitated by accident, neglect, misuse, unauthorized repair or modification ofthe instrument. The instrument may not be returned without a proper Return Authorization Numberfrom Promega, as described below.

This warranty and the remedies set forth herein are exclusive and in lieu of all other express or impliedwarranties (including implied warranties of merchantability, fitness for a particular purpose andnoninfringement), and no other warranties shall be binding upon Promega. In no event shall Promegabe liable for any special, incidental or consequential damages resulting from the use or malfunction ofthis instrument or the system with which it is used.

To obtain service during the warranty period, please take the following steps:

1. Write or call the company that sold you the instrument and describe as precisely as possiblethe nature of the problem.

2. Carry out minor adjustments or tests as suggested by your technical contact.

3. If the instrument is still not functioning properly and needs to be returned to Promega forservice, YOU MUST OBTAIN A PROMEGA RETURN AUTHORIZATION NUMBER prior toshipping the instrument back.

4. Before returning the equipment, you will be responsible for cleaning it and providing aCertificate of Decontamination to Promega in accordance with instructions.

5. After obtaining a Return Authorization Number and signing the Certificate ofDecontamination, pack the instrument carefully (damage incurred in shipping due toimproper packaging is not Promega’s responsibility), write the Return Authorization Numberon the outside of the package and ship it to the address provided by your technical contact.

6. Shipping to and from Promega will be paid by Promega pursuant to directions to beprovided. The instrument will be repaired free of charge for all customers within theirwarranty period.

7. Under no circumstance can equipment be returned without proper authorization. Thisauthorization is needed to ensure that the problem is not a minor problem that can be easilyhandled in your laboratory and to determine the nature of the problem so that repairs can behandled appropriately.

Out of Warranty Service

Contact Promega or your local Promega representative. We will be happy to assist you by telephone atno charge. Repair service, if needed, will be billed at a flat rate to be agreed upon in advance. Yourinvoice will include shipping.
