Paper industry is one of the largest users of starch. Starch is, in fact, the third most prevalent raw material component in paper, only surpassed by cellulose fiber and mineral filler. Depending on the type of paper produced, starch content in the final products may be as high as 10% by paper weight. E' notizia recente che la BASF, in collaborazione con l'olandese AVEBE, sta lavorando allo sviluppo di patate geneticamente modificate al fine di produrre tuberi con livelli maggiori di amido, soprattutto nella tipologia molecolare dell'amilopectina. Queste patate, come la già autorizzata Amflora, aprono interessanti scenari nel settore degli impieghi non-food della fecola di patate.

Un esempio storico consiste nell’uso di una ADP-glucosio PPasi desensitizzata rispetto all’inibitore a feedback 3-PGA (Stark et al., 1992), ma questi risultati non hanno potuto essere riprodotti da un altro gruppo (usando un’altra varietà di patata).
Il sistema modello utilizzato è principalmente la patata.Esamineremo in dettaglio alcuni tentativi di modificazione del contenuto di amido.
Oltre alla composizione (amylose vs. amylopectin) è possibile modificare ed in particolare aumentare la quantità di amido?Diversi gruppi hanno provato ad alterare gli enzimi della via biosintetica. I risultati? Modesti, nel migliore dei casi, o non riproducibili

Risultati non esaltanti (un piccolo aumento nel flusso ad amido) o non riproducibili (nel caso di ADPG PPasi desensitizzata:
A 4–5-fold increase in activity of the enzyme, achieved by transformation with the Escherichia coli ADPglucose pyrophosphorylase gene glgC-16, had no detectable effect on the starch content of developing or mature tubers. No significant effects were found on the contents of ADPglucose, UDPglucose, glucose 1-phosphate, glucose 6-phosphate, PPi, ATP and ADP.

• Strategia sensata nell’economia cellulare perchè se molto troppo ATP viene impiegato per fare amido, la cellula risponde aumentando la respirazione
• Il tubero rischia di andare in ipossia/anossia con tutti i conseguenti pericoli
• Altri transgeni che aumentano il flusso ad amido, se esistono, potrebbero essere molto sensibili alle condizioni colturali (es. terreni fradici)

Evidenze sperimentali che la sintesi di amido può essere limitata dal rifornimento di ATP
* La sovraespressione (o riduzione) del traslocatore plastidiale per ATP/ADP comporta un aumento (o una diminuzione) dell’accumulo di amido in maniera praticamente lineare.
* Incubando fettine di tubero con adenina, si aumenta il pool degli adenilati e anche la velocità di sintesi dell’amido.
ci sono altri enzimi che esercitano un’azione sul metabolismo degli adenilati?

I would also have thought a company would have approached me about the Centeno Plant Cell paper this year but they did not.
