Image: We tested the effects of cellular stress on human CB-MSCs (Neuromics’, SC00A1) since others have reported conversion to the pluripotent state (Heneidi, S., et al., PLoSOne 8: e64752, 2013). We recovered human CB-MSCs from cryopreservation and briefly cultured these cells (48 hours) at 20% O2-5% CO2 in MSC-Gro, low-serum medium (Neuromics, SC00B1). As a stressful environment, these cells were then transferred to 1% O2-5% CO2 at 105/ml in nutrient-poor medium (expired RPMI) supplemented with 0.12 PZ/ml collagenase NB 4 (Serva, catalog number 17454) at 4oC. We retrieved viable cells 48-hours later and transferred them to MSC-Gro humanized, complete medium (Neuromics’, SC00B2). The images above show formation of multi-cell clusters ~50 to 130 um at 2 days (Left) and 4 days (Rt) following exposure to stress. Scale bar is 50 mm.

Following blockage of non-specific binding, cultures were incubated with a Alexa Fluor-conjugated SSEA-3 monoclonal antibody (MC-631-AF 488; BioVision, catalog number 3068-25T) at 10 ml/ml overnight. (B)SSEA-3 positive cells within cell clusters together with propidium iodide. Red cells are non-viable. SSEA-3 is specific to embryonic stem cells; MSCs were negative for SSEA-3.
