The rate of research development at Swinburne again increased in 1994, with the

launching of the Graduate Research School and the announcement of other major

programs, including approval by Council for the establishment of IRIS, our

Industrial Research Institute. IRIS focuses on serving small and medium enterprises 7

throughtechnology transfer, training and resear,c ,, building on th.e University's

successful Centre for Computer Integrated Manu acture.

A high light in late 1994 was the announcement of the success of a joint ind ustry,

university and CSIRO bid for the Cooperative Research Centre (CRC) in International

Food Sc ience and Packaging Technology. Development of the bid was led by staff

of Swinburne's Centre for Applied Colloid and BioColloid Sc ience and CS IRO,

and a report on the aims of the Centre is included in this Report. Swinburne is now

a partner is three CRCs, reflecting our emphasis on co llaborative and applied

researc h of service to the nation.

Much of the University's best-known research is concentrated in four specialist

research centres: the Centre for Applied Neurosciences, Centre for Applied Coll oid

and BioColl oid Sc ience, Centre for Computer Integrated Manufacture (CIM Centre)

and the Centre for Urban and Social Research. All are noted for the ir industry

and community involvement, multidisciplinary approaches and, increasingly,

for internationa l links. At the same t ime, it is pleasing to see the maturing of other

research groups within the University, in f ields which include telecommunications,

information systems, biomedical instrumentation and Asian and international studies.

The University's research output has continued to expand, assisted by substantial increases in external resea rch funding,

including industry support, over the past four years. From 1993 to 1994, Swinburne recorded growth of over 20% in the research

outputs used for ca lculation of research quantum fundi ng from government, testimony to the efforts of staff and students in a

demanding and competitive environment. Research student enrolments are now stabilising after a period of expansion from

a small base. The number of research degrees awarded continues to grow, successful PhD candidates constituting a significant

outcome measure.

Quality assurance is an important feature of research development at Swinburne, and includes measures of cl ient and student

satisfaction. We are implementing a comprehensive Un iversity-wide quality assu rance system, to demonstrate that research and

research supervision are being carried out to the highest standards.

We aim to offer an environment in which our researchers and research students can flourish. To this end Swinburne has invested

in infrastructure to support research growth. Over the next few years, we will complete a substantial program to provide additional

dedicated research space. In 1994, the University acquired prem ises for IR IS and a bui lding to accommodate research students

and staff in Business and Arts. Planning for a Graduate Research Centre will commence in 1995. We aga in acknowledge the

substantial contribution to infrastructure support of our industry partners through funding, in-kind donations and access to external

facilit ies. This Report provides an overview of our research activities. I believe it shows that we are well on the way to achieving our

goal of being a d ist inctive institution, known for qua li ty and user-friendliness in the way we conduct our research.

Swinburne is a key player in the

industry-focused Cooperative Research

Centre (CRC) for International Food

Manufacture and Packaging Science.

This new CRC has generated

substantial industry support for

programs designed to solve technical

impediments to Australian exports

of value-added food products into the

Asian market, which is set to expand to

$160 billion by the year 2000.

Initiated jointly by Swinburne and

CSIRO, this CRC has its office

at Swinburne, with research programs

being carried out at the facilities

of the various partners.

The Ce ntre aims to del iver novel

food-packagi ng processes, products and

systems as well as develop tech nical

and manufacturing knowledge

to enha nce sustainable ma rket growth

and the international competitiveness

of Aust ralia's food and packaging

ind ustries . The research wi ll focus

on three aspects of the food value

ad di ng cha in: food manufactur ing; novel

packaging for life extension;

and package recyc ling and d isposa l.

Advanced packaging mater ials to

be developed by the centre wi ll help

to overcome the chron ic and worsening

waste disposal problems associated with

today's packaging industry, by creating

new, biodegradable and environmentally­

friendly packaging.

In al l, the CRC will attract $30 mi llion

in cash and $48 mi llion in total support

over the next seven years, with industry

providing over 40% of CRC resou rces.

Fielde r, Pac ific Brand Food Group, Pratt

Industr ies, the Horticu ltural Research

$176,000 ARC Collaborative

Research Grant to research and

develop national benchmarks for

social development and citizenship.

The project will address theoretical

and practical aspects of key problems

in Australian social policy development,

including setting goals for and measuring

social well-being.

Development of national indicators and

benchmarks of social well-being in areas

such as children's and youth welfare,

health and disability, housing and legal

rights, will 'help community and

government agencies to more effectively

measure the needs and problems in

their fields', says Research Associate

Michael Salvaris. The results will assist

these agencies in future service delivery

based on international best practice,

and in monitoring national trends

and progress towards the achievement

of these desirable benchmarks.

The . project aims to develop' an '=

Australian social development an_d

citizenship framework including:

an index of gross national well-being

measuring progress in key social policy

fields and a national social agenda,

setting clear targets and benchmarks for

the future. An action plan for practical

application for this framework in

government and the community.

'There must be community involvement

for any recommendations to have

meaning and relevance', says Associate

Professor Burke. 'The support of VCOSS

is critical to the success of the project

and we are grateful for their whole -

Collaborative research into the AIDS virus

is continuing at Swinburne with funding

support from the Commonwealth

AIDS Research Grants Scheme. Chief

investigators John Fecondo, a

senior lecturer in the School of Chemical

Sciences, and Dr Andrew Boyd, of the

Walter and Eliza Hall Institute,

are leading a team carrying out research

into the role of cell surface

adhesion molecules in HIV infection.

Fundamental Studies on Mechanisms of

HIV infection

This research at Swinburne builds

on previous research undertaken

by Mr Fecondo into the role of adhesion

molecules, in particular the ICAM-1

receptor, in the immune process.

Using computerised sequence analysis,

Mr Fecondo has been able to predict which

peptide sequences (amino acids linked by

peptide bonds) are likely binding sites

which block the AIDS virus from getting

into the white blood cells. The peptides are

then chemically synthesised and tested

in trials, with two trials currently under way

at Westmead and Fairfield Hospitals.

While the results are pleasing, Mr Fecondo

stresses the importance at this stage of

pursuing fundamental studies to discover

how HIV penetrates its target cells.

The School of Chemical Sciences has the

potential to carry out further valuable

research in this area with its state-of-the-art

facilities for peptide design, synthesis and

characterisation. Further collaborative

projects are proceeding with institutions

such as the University of Melbourne,

Monash, La Trobe, Flinders and Newcastle

Universities and St Vincents and the

Royal Children's Hospitals.

Industrialisation and

Social Change in Asia

Two decades of research in Asia

by Professor Ken Young has led to many

close and long standing and continuing

collaborative ties with leading academics

in Indonesia, Malaysia, Singapore,

Hong Kong and other parts of Asia.

His work began at London University

and the ANU with studies of social and

economic change in rural Indonesia.

Over the past decade Professor Young

has made significant contributions to the

comparative study of socio-economic

change in the industrialising cities of

Southeast Asia (including Hong Kong).

His continuing comparative work has led to

contributions to a series of books published

by Routledge on "The New Rich in Asia"

prepared by the Asia Research Centre

at Murdoch University. He has also edited,

with Professor R. Tanter, The Politics

of Middle Class Indonesia.

Professor Young's most recent research

investigates industrial isation and social

change in comparative urban contexts,

through a project focused primarily on

Better Viewing through

Interactive Visualisation svsrEMs

Despite the rapid advances

in display technologies in recent

years, one of the major problems

facing computer interface

designers is the relatively small

screen surface available on which

large information spaces need

to be presented.

Research by Dr Ying Leung in the

School of Computer Science and

Software Engineering is looking

at various presentation techniques

to overcome this visual isation difficulty.

More specifically, Dr Leung is

interested in applying distortion­

oriented displays to large information

spaces; the essence of these

techniques is to provide the viewer with

a globa I view of the information space

with local details, thus overcoming

navigat ion and presentation difficulties.

Dr Leung has developed an interactive

visualisation system based on

the bifocal display used for large

topological maps, including the London

Underground map.

Dr Leung is product research leader

in the Swinburne Computer Human

Interaction Labora tory (SCHIU in the

School. His research is supported

by DSTO and has attracted interested

from NEC Japan for its potential

in presenting large computer and

communication networks. Dr Leung

has been invited to spend three months

to further his work at N EC's research

laboratories in Tokyo.

The Swinburne School of Design

added an extra dimension to its work

in 1994 with the appointment of Professor

Allan Whitfield as Research Professor.

This unique appointment reflects a

confidence in the standing of the School's

undergraduate programs and an

expansion into stronger postgraduate

and research provision. 'Designers

have tended not to do research in the

academic sense of the word', says

Professor Whitfield . 'However, research

in the design field has been developing

in ternationally over the past few years and

we are encouraging people from both

a design and non-design background to

pursue research relevant to this field'.

Professor Whitfield came to Swinburne

from the UK, where he was active in the

art and design world before completing

doctoral studies and embarking on an

academic career. For the past fifteen years

he has been funded by the Engineering

and Physical Sciences Research Council

(formerly the Science and Engineering

Research Council) for research in the fields

of colour specification and colour standards.

He has also conducted research into

peoples' perceptions of design and design

management. Professor Whitfield is Cha ir

of a British Standards Committee on

colour standards and also represents the

UK on the ISO Technical Committee which

is attempting to reach international

agreement on how colours are specified.

Dr Romesh Nagarajah

of the University's CIM Centre, had

considerable experience in the

manufacturing industry before taking

up a career in academia in 1985.

His pioneering work in the application of

robotics to crystal glass manufacture led

to his masters degree from the

University of Nottingham, while his PhD

work resulted in the development

of a methodology for cell sizing in flexible

manufacturing systems based on studies

of the manufacturing characteristics

of a number of companies involved in

discrete manufacturing.

Dr Don Phillips

PhD evolved from his extensive work

on-site stormwater detention (OSD) with

industry ·and municipal councils. OSD

is used to relieve the problems of flash

flooding in urban areas, an occurrence

which has been exacerbated by the

growing trend towards urban consolidation

and has resulted in serious property

damage. A senior lecturer in the School

of Civil Engineering and Building,

Dr Phillips' work has led to the widespread

adoption of his design procedures and

to patents for effective outflow control

devices in OSD and subsequent

manufacturing licences in Victoria and

New South Wales.

Dr Eric Woxvold

an engineering science graduate of the

US Naval Academy at Annapolis,

Maryland, has spent almost three decades

implementing and developing

manufacturing management software

systems. During his research in

the University's CIM Centre, Dr Woxvold

developed a unitary structure (ARMOR)

that enabled the project management,

production control and manufacturing

resources planning of complex devices

to be undertaken within a single software

system. Software based on Dr Woxvold's

original research is now used in a number

of defence assembly and maintenance

applications.

Dr Seung-Rok Kim

working in the Centre for Applied Colloid

and BioColloid Science, completed his

thesis on the flow and stability of

nonaqueous dispersions. His main study,

on the behaviour of particles dispersed

in oil, is directly applicable to industry in

fields such as to oil refining, papermaking

and the manufacture of ceramics.

Dr Andreas Pitsillides

of the School of Biophysical Science and

Electrical Engineering, is working on

various research issues in the automatic

management and control of

telecommunication networks. His research,

looking towards the 21st century, has an

emphasis on ATM-based multiservice and

multimedia networks. Dr Pitsillides' thesis

focused on the regulation of network user

traffic and the efficient allocation of

network resources to users while satisfying

service quality requirements.

Mossbauer spectroscopy, discovering

processes of precipitation and absorption

of heavy metals into colloidal surfaces, with

application to industry and the environment.

condensation inside the microporous solids,

a more accurate microporous absorption

Lambie, D. & Clarke, S. R. An Investigation into the Mathematical Modelling of the Dynamics of the Lawn Bowl To predict performance characteristics and

automating bowl design.

Cadusch, P.J. An Investigation of Methods to Interpolate and Invert Topographic EEG Data to Handle more Realistic Head Geometries and Conductive Distributions A technique based on properties of spherical

splines is currently used for deblurring

topographic EEG data to remove the effects

of volume conduction. This scheme is

somewhat limited by the assumption of

spherical symmetry of the conductivity

distribution. An extension of this scheme,

using spline-type fitting based on a finite

element method, is proposed and currently

under investigation.

Cadusch, P.J. & Silberstein, R.B. Development of a New Technique, Using Time-frequency Signal Analysis, for Seg­menting EEG Time Sequences on the Basis of Changes in the Topography of the Scalp Recorded Data Results suggest that previous approaches

based on topography alone may be overly

sensitive to small (possib ly artifactual}

variations in the data .

Hendtlass, T. Artificial Neural Networks with Problem Dependent Architectures An adaptive neural network has been

developed that is capable of distinguishing

between voca l utterances over short t ime

intervals. The network utitilses exemplar

tracking ard exemplar substitution approaches.

Nunez, P.L., Silberstein, R.B._, Cadusch, P.J., Wijesinghe, R., Westdorp, A. & Srinivasan, R. Localization of Brain Activity Two different methods to improve the

resolution of topographic EEG have been

compared; one based on surface Laplacians

developed by the Brain Physics Group at

Tulane University and the other based on

spatial deconvolution using spherical splines

developed at SUT. Both methods perform

well with simulated data and produce

similar results with experimental data.

Sergejew, A.A. An Investigation of the Pattern Recognition Capabilities of Artificial Neural Network Methodologies to EEG Signals Recorded from Subjects with Serious Psychiatric Disorders This long-term project has the potential for

produc ing EEG-based biological markers

of information processing disturbances in

a variety of diagnostic groups.

Sergejew, A.A. & Tsoi, A. To Complement Traditional Topographic EEG Analysis Techniques by Capturing Instantaneous EEG Signal Dynamics in Obsessive-compulsive Disorders using Markov Modelling Techniques to Quantify State Transition Rules Preliminary results have confirmed our

hypothesis regarding the type of thought

disorder comprising OCD.

Silberstein, R.B, Ball, J. & Currie, J. An Investigation of the Sensitivity of Steady State Visually Evoked Potentials to Localised Brain Damage Using Probe-ERP techniques, with an

attentional task, any changes in evoked

potentials as a result of brain damage will

be investigated and reported. The aim is to

relate the structura l changes (caused by

brain damage} with atientional processes.

Silberstein, R.B & Chant, G. Mass Communication and Visually Evoked Potentials This project involves the application of

Steady State Probe Topography (SSPT} for

the assessment of viewer interest in visua lly

presented material. This project has

significant applications in the Market

Research industry.

subjects while they repetitively perform a

"automatic" and effortless. A principle

feature of this project is the changes in

brain activ ity during the process of

automatization.

Ascorbate as a Preferred Substrate for

Peroxidase

Hydrogen peroxide is an important oxidising

agent in mammalian systems. Metabolically

effic ient disposal of this agent would help

delay metabolic dangers due to aging.

This project will examine the interaction

of ascorbate with hydrogen peroxide

catalysed by peroxidases which may prove

to be an efficient mechanism for disposal

of peroxidase.

Ascorbic Acid Analysis and Disulphide

Groups

In the presence of copper ion ascorbic

acid produces hydrogen peroxide. Certain

proteins, wh ich are known to have exposed

disulphide groups, also interact with

ascorbate and copper and the extent of

interaction can be related to the standard

curve prepared from varying concentrations

of cystine. These proteins also affect the

production of hydrogen peroxide from

ascorbate at biological levels of copper ion.

Ascorbic Acid and other Electron Donors

Haemoglobin, myoglobin and catalase show

peroxidatic activity. A number of specific

electron donors, which inc lude ascorbate,

Peroxidase Activity

Horseradish peroxidase acts as a catalyst

in the transfer of electrons from a donor

to reduce hydrogen peroxide. The Ellman's

reagent anion has been shown to act as

a donor with loss of colour and oxidation

occu rs in the Ngo-henhoff reaction which is

the most sensitive colorimetr ic reaction for

perox idase. Ascorb ic acid has been shown

to prevent the ox idation of the above donors

in the presence of horseradish peroxidase.

receptor. The secreted protein appears less

heterogeneous than that produced from

recombinant CHO ce lls and may be more

Expression System

Apolipoprotein Al (ApoAI) is the principal

protein component of high density

lipoproteins (HOU which have a protective

role aga inst atherosclerosis by clearing

cholesterol from the system. ApoA I is

synthesised in hepatic and intestina l cells

as a preproprotein and secreted as a

proprotein (proApoAI) upon removal of the

signa l peptide. A further six amino acids are

removed from the N-terminus to generate

the 28 kD ApoAI in the serum. Stud ies on

ApoAI structure and function are hampered

by the difficulty in extracting ApoA I away

from serum proteins and lipids. To produce

the protein in a serum-free expression

system the cDNA fo r the gene was

engineered into a baculovirus and insect

ce ll s grown on serum-free med ium were

infected with the recombinant virus.

A Multidomain Precursor Protein that is

Processed into 5 Homologous Proteinase

Inhibitors in the Stigma of Nicotiana alata

A cDNA corresponding to a gene for a 43 kD

serine protease inhibitor (Pl) precursor has

been isolated from Nicotiana alata (tobacco

plant). The precursor is expressed in the

stigma, where it appears in the vacuole,

and is processed by proteolytic cleavage

into five homologous protease inhibitors of

6 kD. One of the five inhibits trypsin while

the other four inhibit chymotrypsin.

These protease inhibitors interfere with the

digestion of protein in the gut of caterpillars.

In order to obtain enough protein to study

the structure and processing of the protease

precursor the cDNA was inserted into a

recombinant baculovirus and cultured insect

cells were used to produce the protein.

The protein produced by infected insect

cells was secreted into the medium and its

N-terminal sequence (KACTLN) was identical

to the plant precursor showing that the

signal sequence was functional in insect

cells and was cleaved correctly. The precursor

from insect cells had chymotrypsin inhibitory

activity but no activity towards trypsin until

it was digested with the protease AspN,

releasing the 6 kD peptides.

Expression by Insect Cells of MOG,

a Major Autoanigen of Multiple Sclerosis

Myelin oligodendrocyte glycoprotein (MOG)

is a transmembrane protein expressed only

in the central nervous system. It is one of

the major autoantigens of multiple sclerosis

(MS). To carry out studies of T-lymphocyte

interaction with MOG it is necessary to

produce the protein in a system where there

are no other human proteins and no bacterial

products that could contaminate the purified

protein. The cDNA for MOG has been placed

in a recombinant baculovirus. MOG in

infected insect cells has been identified by

immuno-blotting. Purification of the protein

from cultures will be carried out to produced

material for an analysis of MS patient

T-lymphocyte response.

Binding Proteins Involved in Autoimmunity

The nuclear autoantigens La, Ro60

and Ro52 are components of the

ribonuc leoprotein (RNP) complex in RNA

transcription. These proteins become the

target of autoimmune responses in several

common autoimmune disorders. This

project aims to produce these proteins in

the baculovirus expression system to enable

a study of their interaction with T-lymphocytes

and antibodies. The cDNA for La (a 47 kD

phosphoprotein) has been engineered into

a recombinant baculovirus and infected

insect cells produce large amounts of the

La protein. It is intended to purify enough

protein in to enable crystallographic studies

to be carried out.

fluorescent adducts of glutathione when

treated with glutathione-S-transferase.

This reaction is now being investigated

as a sensitive assay method for reduced

glutathione.

Inhibitors and Sulphonamides by Capillary

Electrophoresis and HPLC

Separations of these two classes of

drugs are examined by capillary zone

electrophoresis. Salt effects have been

examined and molecular modelling used

to verify the fundamental nature of the

separation mechanism. The DHFRI have

also been separated by HPLC and further

molecular modelling is intended in this

context. Separation of both classes of drugs

will also be investigated via micellar

electrokinetic capillary chromatography

(MECC) and procedures optimised via

mathematical procedures.

The GABA receptor is a supramolecular

cological responses. Molecular modelling

involving the definition of one binding site

study of the predicted secondary structure

of one protein subunit is being used to

develop a pharmacophore model which will

assist in the development of novel agonists

which might bind with partial occupancy at

the same site. Several molecules are shortly

Structure-Function Studies of lntercellular

Adhesion Molecule-! (ICAM-1)

We and others have shown that ICAM-1 has

a central role in a wide variety of adhesion­

dependent leucocyte functions including

T and B cell activation, leucocyte migration,

inflammation; and also has been a role in a

number of disease states includ ing malaria,

arthritis, asthma and the common co ld.

Given its central role in the immune system

and in a number of disease states, we have

attempted to define the functional binding

sites within the ICAM-1 sequence with a

view to developing small peptides as

inhibitors of ICAM-1 -mediated adhes ion.

We have delineated two regions of funct ional

importance within domains 2 and 4 of ICAM-1

and have shown that synthetic peptides

based on these sequences (JF9 and JFlO)

mimic the effects of our anti-lCAM-1

monoclonal antibody, WEHI-CAM-1.

The primary aim of this project is to explore

structure-function relationships of the

cytoplasmic domains of two B lymphocyte

cell surface molecules that belong to different

receptor families, namely: (a) lntercellular

Adhesion Molecule-1 (ICAM-1, member of

lgSF} and (b) Human eph-like kinase (hek,

belongs to RTK family) . The general strategy

being used initially is the preparation of

"affinity chromatography" columns that

contain synthetic forms of the cytoplasmic

domains of these receptors to isolate cyto­

plasmic and/or membrane proteins from cell

lysates that may associate with the receptors.

Any cytoplasmic components that are isolated

will be subjected to a detailed analysis using

a variety of immunochemica l and protein

chemistry methods. Further studies will

include identification of the functional role(s)

of the binding proteins and thus provide

further insight into some of the underlying

mechanisms involved in the signal transduction

processes that involve these molecules.

To develop convenient routine methods to

identify and analyse trace levels of nony­

lphenol ethoxylates in environmental

samples. Th is class of non-ionic surfactants

is extensively used in industry, for example,

wool scouring. They, and even more so

their breakdown products, are toxic in the

environment. HPLC methods of identification

and analysis have been developed. The

assignment of the HPLC chromatogram has

been confirmed by GC and GC/MS. Various

methods of extracting and preconcentrating

the surfactants have been evaluated and

gas stripping into ethyl acetate selected .

The overall method has been applied to the

analysis of a number of sewage effluents.

It has been adopted as an official EPA

method and is currently undergoing inter­

laboratory trials with CSIRO, Division of Wool

Technology.

Conformational Analysis of Antipsychotic

Drugs using NMR and Computational

Chemistry Techniques

Antipsychotic drugs act at a variety of neuro­

transmitter systems but to different extents .

A series of atypical antipsychotic drugs will

be analysed both by NMR and computational

chemistry methods to establish the possible

shapes of these molecules. The range of

individual molecular shapes will be compared

to determine if they have enough chemical

properties in common to explain their precise

pharmacological actions. This information

will be valuable in understanding both their

mode of action and in the design atypical

antipsychotics devoid of side effects.

Building Regulations may require building

elemen ts to be insu lated aga inst fire. The

requirements for determi ning the insu lation

material and thickness are currently based

on conservative regulatory codes of practice

based on past experience. These 'rules of

thumb' requ irements va ry from country

to country. The ai m of this research is to

investigate the f ire req ui rements at an

international leve l and to develop sc ienti fic

requ irements for fire rat ing build ing elements

in an economical cost effective way.
