In the context of host HLA molecules, human T cells recognize foreign antigens through the T-cell receptor (TcR). The TcRs are molecular complexes comprised of two units: a recognition unit, composed of either a-b or γ-δ heterodimer (present on the cell surface in a mutually exclusive manner) and a transducing unit referred to as the CD3 complex. This complex which is commmon to a-b and γ-δ heterodimers, triggers the T cell when the recognition unit is occupied by the antigen. The recognition unit’s function is to recognize any possible foreign antigen; the diversity necessary for this function is generated by somatic recombination of the TcR genes. There are four TcR gene loci (a, b, γ and δ). Each is composed of several V (for variable) segments, coding for about 90 amino acids, very short D (for diversity) segments for a and δ loci only, and short J (for joining) segments of approximately 15 amino acids and one or two C (for constant) segments.
