Youhavepurchasedthestate-of-the-artinprecisionpipetting.TheThermoScientificMatrixElectronicPipetteisthefirstcordlesselectronicpipettethat“understands”themyriadofliquidhandlingprotocolstypicallyperformedintoday’slaboratory.

Ergonomicallydesignedandbalancedtoreducefatigue,theMatrixelectronicpipettefunctionssointuitively,youmayneedonlyaquicksurveyofthismanual to use it.

Theelectronicpipetteexecutesyourliquidpipettingtasksquickly,accurately,andwithlessriskofdevelopingrepetitivestraininjuries(RSI)typicallyassociatedwithmanualpipetteuse.Laboratoryresultswillbecomemorestandardized,whilelesstimeisexpendedontediousbenchprocedures.

Before You Begin

A. INTRODUCTION

TheThermoScientificMatrixElectronicPipetteprovidestheconvenienceofamanualpipettor,withtheprogrammingcapabilitiesofanelectronicpipette.TheMatrixelectronicpipettefeaturesinclude:

•Expandedmemoryforstoringuptofivepipettingprograms.

•Eachprogramcanholdamaximumof40steps.

•“ScratchPad”programmingmodewhichallowsforfastandsimpleprogramchangesthatarenotstoredintomemory.

•Enhancedspeedcontrolofferssloweraspiratinganddispensingofviscousliquids.Ideal for gel loading!

•“EASY”CALwhichsimplifiesin-labcalibrationverificationusing distilledwater.

TheMatrixelectronicpipetteislightweightandcordless,utilizingrechargeablebatteries,thatcanbeusedduringrecharge(with the exception of single channel pipettes).Thepipettealsohasanautomaticshut-offfeaturetoextendbatterylife.

Theelectronicpipetteusesintegratedcircuittechnologytostoreprogramminginformation.Itwillholdupto40stepsinaprogram,withupto5programssavedinmemory,and1scratchpad(See section C.1.1 Open Programming Mode or “Scratch Pad”).Theprograminformationisretainedinmemoryevenifthebatteriesarefullydischargedorremoved.

Theelectronicpipette usesa“paceddispense”featurethatperformsrepeatpipettingstepswhilethetriggerisdepressed.

Thesinglechannelelectronicpipetteareavailableinanumberofmodelconfigurations(see Figures A-1)from12.5µlto1250µl,ranginginvolumefrom0.5µlto1250µl.Eachpipetteiscolor-codedforeasyvolumeidentification(see section B.5, Pipette Specifications).Twomodels,250µland1250µl,areofferedwithashorterejectorsleevereducingtheoveralllengthcomparedtothestandardpipette.Idealforuseinconfinedspacessuchasbiosafetycabinet

B.2 General Description

Keypad.Thekeypadcontainstheoperationkeysthatareusedinprogrammingthepipettingsequenceandvolumes. (See Section B.3, Keypad).

Display.ThedisplayisanLCDscreenthatshowsthepipettingoperation(fill, dispense, mix, or purge)andfluidvolumeforeachstepinthepipettingprogram.(See Section B.4, Display).

Battery Case.Thebatterycasecontainstherechargeablebatteries.NickelMetalHydridebatteriesfullychargein14-16hours,whileLithium-Ionbatteriesfullychargein2hours.Thepipettecanperformafullday’sworkloadwithnoneedtorecharge.Batterylifeisextendedwithautomaticshut-offafter 10minutesofinactivity.(See Section F.3, Installing/Replacing Batteries).

Pipetting Trigger.Thepipettingtriggerinitiatesaprogram.Asinglepipettingstepisexecutedbyaquickpressandreleaseofthetrigger.Repeatpipettingisexecutedbypressingandholdingthetrigger.

Note: At the end of any dispense, the pipette tips should be removed from the fluid before releasing the trigger, to prevent aspiration of fluid.

Ejector Trigger.Theejectortriggerispressedtoejectthetips fromthepipette.

Tip Fittings.Thetipfittingsformanairtightsealwiththetipstoensurepipettingaccuracy.

Note: Electronic pipettes are equipped with chemically-resistant (PEEK) tip fittings (and ejector sleeves on single channel models) that allow pipetting of acidic solutions without damage to the tip fittings.

Color-coded Ring.Theringaroundthepipettehandleiscolorcodedtoidentifythepipettingvolumeofthehandle(See Section B.5, Pipette Specifications).

Transformer Connector.Thetransformerconnectorreceivestheplugfromthepowertransformerforrechargingthebatteries.Thepipettecanbeusedwhilerecharging(with the exception of single channel pipettes).

B.3.1 Operation Keys

[On/Off] TheOn/Offkeyturnsthepipetteonoroff.Whenthepipetteisturnedon,itemitsabeeptone,flashes“ZERO”onthedisplay,thenautomaticallyresetsthepistonstozeroposition.Afterzeroing,thedisplayshowsthefirststepoftheprogramintheRUNmode.

Note: If the pipette is being programmed for the first time, the first step will be the fill operation, with the default fill volume. (See Section C.1, Programming the pipette, for the default settings).

[Prog] TheProgramkeyontheMatrixelectronicpipetteenablesyoutostoreuptofiveseparateprogramsinmemoryforfuturerecall.Eachprogramcanholdupto40steps.Programsarestoredinreservedprogramlocationnumbers1–5.Program“0”isusedasa“scratchpad”wheretemporaryprogramscanbewritten,run,andoverwritteneasily.Program“0”isconsideredtobean“open”program.(See Section C.1, Programming the Pipette).

[Purge] ThePurgekeyentersapurgepromptanytimeduringapipettingoperationtoaborttheprogram.BypressingthePurgekey,thenpressingthetrigger,youcandispenseanyremainingliquidfromthepipettetipsandreturntothebeginningoftheprogram.

ThePurgekeyisalsousedtoenterthepurgestepintoaprogram.

Note: If there is insufficient liquid for a final dispense step, the pipette emits a beep tone and automatically displays the purge prompt.

Note: The RUN mode performs the operation that is displayed, such as fill, dispense, mix, or purge. When “Run” appears in the display, you can activate the RUN mode by pressing the trigger. At the end of a pipetting sequence, the pipette emits a triple beep tone and returns to the beginning of the program. The pipette remains in RUN mode until you program a new pipetting sequence.

TheFill,Dispense,andMixkeysprogramthepipettingoperations.Bypressingthefillormixkey,youcanentertheprogrammingmodeoftheelectronicpipette.The“Run”promptdisappearsfromthedisplayandtheappropriateoperationappearswithaflashing“Vol”prompt.Fill,dispense,ormixvolumescanbeprogrammedbypressingtheVolumekey.Also,thenumberofmixcyclescanbeprogrammedbypressingtheMixkeytwice.

Automatic blowout:Attheendofafinaldispensestep,thepipetteautomaticallyexpelsanyremainingliquidfromthepipettetipswithashortburstofair(blowout).Thepistonsthenreversedirectionandreturntozeroposition.

TheVolumekeycontrolsthequantityofliquidtobeaspiratedordispensed.Thevolumecanbeenteredwhenthe“Vol”promptappears.

PipettingvolumecanbeincrementedquicklybycontinuouslydepressingtheVolumekey.Thelongeryouholdthekey,thegreatertheincrementalspeed.

TheVolumekeycanalsobeusedtochangepipettingspeedswithinastoredprogramorduringtheexecutionofaprogram.(See Section C.1.5, Adjusting Pipetting Speed).

[Enter] TheEnterkeystorestheprograminmemory.Aftereachstepisprogrammed,theEnterkeyispressedoncetoacceptthestep.Afterthefinalstepisprogrammed,theEnterkeyispressedtwicetodenotetheendoftheprogram.Thepipetteverifiestheendoftheprogramwithatriplebeeptone.

B.3.3 Special Function Key

TheSpecialFunctionkeyisthesmallholebelowtheVolumekey.Itcanbeactivatedwithapipettetip.ByrepeatedlypressingtheSpecialFunctionkey,youcandisplayfourfunctions:PipettingSpeed(SPd),BeepTone(TONE),PacedDispenseSpeed(PACE),andCalibration(CAL).ToexittheSpecialFunctionkeyatanytime,presstheEnterkey.

PACE Thepacecontrolsthespeedofrepeatpipetting(paced dispensing),FAST,SLOWorOFF.PresstheSpecialFunctionkeythreetimestodisplay“PACE”anditscurrentsetting.Tochangethesetting,presstheVolumekey.

PacedDispense.Whenusingthepaceddispensefeature,thepipettedispensestheprogrammedvolumeatthespecifiedpace(fast or slow).Immediatelyaftereachdispensestep,thepipetteemitsasinglebeeptone(if beep tone is on)toprepareyouforthenextdispensestep(see Section C.5, Operating Procedures).

CAL Thecalibrationofthepipetteisfactorysetfordistilledwateratroomtemperature.Ifyouarepipettingfluidswithsignificantlydifferentspecificgravitiesortemperatures,usethissettingtorecalibratethepipette.(See Section D, Calibration).

Thecurrentspeedsettingappearsatthebottomofthedisplay.Thesesettingscanbeselectedandchangedatanytimebeforeaprogramstepisexecuted.

C.1 Programming the Electronic Pipette

ProgrammingtheMatrixelectronicpipetteisfastandeasy.Pipettingstepsandvolumeareenteredinthesequencethattheywillbeperformed,thentheprogramissavedtomemory.Thisprogramwillstayinmemory(evenifthepipetteisturnedoff,ortheelectronicpipettebatteryrunslow)untilyouchangeit.Fiveprogramsandonescratchpad(Program0),eachcapableofholding40steps,canbeenteredandstoredintheelectronicpipette.

C.1.1 Open Programming Mode or “Scratch Pad” (Program 0)

Purge.The[Purge]keyprogramsthepipettetopurgethefluid.Thisstepcanbeenteredasaprogramstep,oritcanbepressedanytimewhilepipettingtoterminatetheprogramanddispensetheremainingliquid.

Note: If the program ends at a volume greater than zero, and the last step is not a mix cycle, the “Purge” prompt automatically appears. Also, the pipette emits a double beep tone to warn you that some fluid remains. Press the trigger to dispense the remaining fluid and return to the beginning of the program.

• Pipette at a constant temperature. Allowliquidsandequipmenttoequilibratetoambienttemperature.Volumeaspirationwithairdisplacementpipettesvarieswithairpressure,relativehumidityandtheliquidsvaporpressure,allofwhicharetemperaturedependent.

• Attach tips with slow, even pressure.Whenapplyingtips,avoid‘banging’orapplyingexcessiveforcetothetip(s),asthisstretchesthetipsealandwillaffectvolume.UseMatrixtips,whichhavebeenspecificallydesignedtomatchthetipfittingsonMatrixpipettes.

• Calibrate the pipette for the liquid.Iftheliquidhasaspecificgravityandvaporpressureequivalenttodistilledwater,calibrateusing“EASY”CALmode.Otherwise,use“CALC”mode.

• Pre-wet the pipette tip.Aspirateanddispenseafewtimesbeforeaspiratingtheliquidtoincreasehumiditywithinthetip,reducingsampleevaporationthatresultsinlowerdispensevolumesininitialfewpipettings.Theneedtopre-wetincreaseswhenworkingwithliquidswithhighvaporpressureand/orhighsurfacetension.

• Immerse tips fully.Tipsshouldbeimmersedapproximately1/4inch(approx. 0.6 cm)belowtheliquidsmeniscusandawayfromthecontainersurfacestoavoidaspirationofairandexcessdropletsclingingtotheoutsideofthetip.

• Discard a primer volume.Whenthemotorchangesfromaspirationtodispense,thepercentageoferrorisgreaterinthefirstdispensethaninsubsequentdispenses.

• Pause consistently.Ittakesamomentfortheliquidinthetiptofinishmoving.Pausewiththetipintheliquidforaboutonesecondafteraspiratingthesampletoallowtheliquidtofinishmoviingintothepipettetipafterthepipettestops.Otherwise,thevolumewillbelowerthanitshould.

• Aspirate slowly, dispense quickly.Asageneralrule,pipetteliquids,especiallyviscousliquids,ataslowerspeedtoensureaccuratepipetting.Dispenseaqueoussolutionsatfasterspeedstoexpelliquidmore accurately.

Note: Viscous liquids should be dispensed slowly.

• Tip touch.Duringthedispensingofaliquid,touchthetipagainstthesideofthereceivingcontainertopreventanyliquidfromremainingonthetip.

• Discard final dispense.Whenaccuratedispensingofseveralincrementsofliquidareneeded,discardthelastincrement.Thecumulativepercentageoferrorfrompreviousincrementscarriestothelastvolume.(i.e. The pipette aspirates 100 µl and dispenses 20 µl five times, and if each of the first four dispenses were off by +1.0 µl, then the last dispense is missing 4 µl before you even dispense it).

5. Insert new battery pack. Insertthenewbatterypackintothecavityofthemainhousing.Ensurethatthewireisloopedunderthebatterypackandtheconnectorisaboveit(refer to Figure F.3.1Ac).

NOTE: The battery pack fits very snugly in the main housing; therefore, ensure that the pack is oriented as shown in Figure F.3.1Ac. The connectors for the battery pack and the pipette

should be on opposite sides.

6. Reconnect wire connectors. Reconnectthewireconnectorsabovethebatterypack.Gentlypushthewiresofthebatterypackdownuntilthewireconnectorslayflatoverthetopofthebatterypack.

TheelectronicpipetteusestwoNickelMetalHydride(NiMH)rechargeablebatteries,locatedinabatterycaseunderthekeypad.Thebatteriesarechargedinthepipettebyusingthepowertransformerincludedinthepackage,withtheexceptionofsinglechannelpipetteswhichdonothavethisability.Youcanoperatethepipettewhileitisrecharging,oryoucanoperateitsolelywiththepowertransformer(withoutbatteries).Thebatteriescanberechargedapproximately400-500times.Toextendbatterylife,thepipettewillturnoffautomaticallyifitisnotusedwithintenminutes.

1. Attach transformer.Attachthecableendofthetransformerintothepipette.Plugthetransformerintotheelectricaloutlet.

2. Recharge batteries.Rechargebatteriesfor14-16hourstoensureafullcharge.Afterthebatterieshavebeenrecharged,unplugthepipettefromthe transformer.

Note: Batteries hold their maximum charge if they are fully discharged before recharging again. However, overnight recharging is acceptable if the pipette is in normal use during the day.

Note: The typical life expectancy of the battery is approximately 400-500 full recharges. To prolong this and attain the maximum usage from the battery, it is recommended the battery be recharged whenever it is not being used instead of waiting until the battery is completely discharged. Completely draining the battery will not shorten the life expectancy. However, recharging before fully discharged will prolong the battery life.

CAUTION! Avoidexcessivelychargingthebatterieswhenthepipetteisnotinuse(excluding16channelpipettes).Excessivechargetothebatterieswillshortenthebatterylifeandmaydamagethebatteries.
