© Procainamide and napa immunogens, antibodies labeled conjugates, and related derivatives. Procainamide and N-acetylprocainamide (NAPA) im- munogens, antibodies prepared therefrom, labeled conju- gates, synthetic intermediates, and the use of such anti- bodies and labeled conjugates in immunoassays for deter- mining the respective drugs. The immunogens comprise the drugs coupled at the a-position of the amide side chain to an immunogenic carrier material. The labeled conjugates and synthetic intermediates similarly are a-position derivatives of the drugs or precursors thereof. The antibodies and labeled conjugates are particularly useful in homogeneous nonradioisotopic immunoassays for measuring the respec- tive drugs in biological fluids such as serum.

BACKGROUND OF THE INVENTION

1. FIELD OF THE INVENTION

T h i s i n v e n t i o n r e l a t e s to n o v e l p r o c a i n a m i d e a n d

N - a c e t y l p r o c a i n a m i d e (NAPA) d e r i v a t i v e s p e r t a i n i n g t o

i m m u n o a s s a y s f o r d e t e r m i n i n g s u c h d r u g s in l i q u i d

m e d i a s u c h as b i o l o g i c a l f l u i d s . The d e r i v a t i v e s

i n c l u d e i m m u n o g e n s u s e d to s t i m u l a t e p r o d u c t i o n o f

a n t i b o d i e s to t h e d r u g s in h o s t a n i m a l s by c o n v e n -

t i o n a l t e c h n i q u e s . Also p r o v i d e d a r e l a b e l e d c o n j u -

g a t e s u s e d as r e a g e n t s , a l o n g w i t h t h e a n t i b o d i e s ,

in p a r t i c u l a r l y p r e f e r r e d i m m u n o a s s a y s . I n t e r -

t h e t h e r a p e u t i c u s e f u l n e s s and t h e t o x i c i t y o f

t h e d r u g s a r e b e t t e r c o r r e l a t e d w i t h t h e i r b l o o d

l e v e l s t h a n w i t h t h e i r d o s a g e s . The r e l a t i o n s h i p

b e t w e e n t h e a m o u n t of d rug a d m i n i s t e r e d and t h e

by c o m p l e t e n e s s of a b s o r p t i o n , d i s t r i b u t i o n c h a r a c -

SUMMARY OF THE INVENTION

The p r e s e n t i n v e n t i o n u n i q u e l y p r o v i d e s r e a g e n t s f o r u se in p r o c a i n a m i d e and NAPA i m m u n o a s s a y s i n v o l v -

ing t h e c o u p l i n g to or d e r i v a t i z a t i o n of t he d r u g

a t t h e a - p o s i t i o n on t h e amide s i d e c h a i n . T h e

i m m u n o g e n of t h e p r e s e n t i n v e n t i o n , c o m p r i s i n g t h e

h a p t e n i c d r u g c o v a l e n t l y l i n k e d a t t h e a - p o s i t i o n t o

an i m m u n o g e n i c c a r r i e r m a t e r i a l , s t i m u l a t e s t h e

p r o d u c t i o n of a n t i b o d i e s to t h e r e s p e c t i v e d r u g s . By

c o u p l i n g t h e d r u g at t h e a - p o s i t i o n w h e r e no s u b s t i -

t u e n t s a p p e a r in t h e p a r e n t d r u g s , t h e immunogen c o n -

j u g a t e is p r e p a r e d w i t h o u t m o d i f y i n g any f u n c t i o n a l

or d i s t i n g u i s h i n g g r o u p s on t h e d r u g s .

In a p r e f e r r e d e m b o d i m e n t , t h e p r e s e n t i n v e n t i o n

p r o v i d e s n o v e l i n t e r m e d i a t e s in t h e p r e p a r a t i o n o f

t h e a - s u b s t i t u t e d d r u g - c a r r i e r i m m u n o g e n s . A l s o

p r o v i d e d a r e an i m p r o v e d i m m u n o a s s a y m e t h o d a n d

r e a g e n t means f o r t h e d e t e r m i n a t i o n of t he d r u g s

w i t h t h e u se of t he n o v e l a n t i b o d i e s of t h e p r e s e n t

i n v e n t i o n . The p r e s e n t i n v e n t i o n a l s o p r o v i d e s

l a b e l e d c o n j u g a t e s f o r p a r t i c u l a r l y p r e f e r r e d e m b o d i -

m e n t s of s u c h i m m u n o a s s a y .

t i o n was a d d e d 20 g of a n h y d r o u s K2CO3 and 7 .14 g ( 3 8

mmol) of 4 - n i t r o b e n z o y l c h l o r i d e . A f t e r s t i r r i n g f o r

12 h o u r s at room t e m p e r a t u r e , s o l v e n t was r e m o v e d u n d e r

r e d u c e d p r e s s u r e , and t he r e s i d u e p a r t i t i o n e d b e t w e e n

e t h e r and 10% NaOH s o l u t i o n . The o r g a n i c p h a s e w a s

s e p a r a t e d , w a s h e d w i t h a d d i t i o n a l 10% NaOH s o l u t i o n ,

1 h o u r a t 0°C. E x c e s s t r i f l u o r o a c e t i c a c i d was r e - moved u n d e r r e d u c e d p r e s s u r e and t h e r e s i d u e was t a k e n

up in H20. I t was f i l t e r e d and t h e f i l t r a t e s a t u r a t e d

w i t h KC1, and made b a s i c w i t h NaOH. The s o l u t i o n w a s e x t r a c t e d w i t h CHCl3 and t h e e x t r a c t d r i e d , f i l t e r e d ,

and e v a p o r a t e d . T h i s l e f t 1 .4 g of t h e a m i n o -

f u n c t i o n a l i z e d p r o c a i n a m i d e d e r i v a t i v e ( 3 ) , Z = NH2, n = 4 as a c l e a r o i l .

B. P r e p a r a t i o n of i m m u n o g e n

The a m i n o - f u n c t i o n a l i z e d p r o c a i n a m i d e d e r i v a t i v e

p r e p a r e d a b o v e [9 .5 mg (35 µ m o l ) ] was d i s s o l v e d i n

0 .5 mL of d i s t i l l e d w a t e r in a t e s t t u b e . A s m a l l

m a g n e t i c s t i r b a r was a d d e d and t h e pH of t h e s o l u t i o n

was a d j u s t e d to pH 4 .0 w i t h 0 .1 n o r m a l (N) HCl. T h e

s o l u t i o n was c o o l e d in an i c e b a t h and 9 .6 m i l l i g r a m s

(50 µ m o l e s ) of l - e t h y l - 3 - ( 3 - d i m e t h y l a m i n o p r o p y l )

c a r b o d i i m i d e was a d d e d a n d d i s s o l v e d . B o v i n e s e r u m

a l b u m i n (25 mg, 35 µ m o l e s ) d i s s o l v e d in 0 .5 mL o f

d i s t i l l e d H2O was s l o w l y a d d e d t o t he s o l u t i o n c o n t a i n -

ing t h e p r o c a i n a m i d e d e r i v a t i v e and c a r b o d i i m i d e .

The r e a c t i o n i n c u b a t e d f o r 24 h o u r s at 4°C, and t h e n

was d i a l y z e d a g a i n s t one l i t e r of d i s t i l l e d w a t e r w h i c h

was c h a n g e d t h r e e t i m e s . Sodium a z i d e was a d d e d to a f i n a l c o n c e n t r a t i o n of 0 .1%. The p r o t e i n c o n c e n t r a t i o n

of t h e immunogen was a d j u s t e d to a c o n c e n t r a t i o n o f

1 mg/mL w i t h s a l i n e (0 .85% N a C l ) .

Six m i l l i l i t e r s of immunogen (1 mg/mL) was com- b i n e d w i t h 12 mL of F r u e n d s C o m p l e t e A d j u v a n t a n d

6 mL of s a l i n e . R a b b i t s were i m m u n i z e d s i m u l t a n e o u s l y e a c h w i t h 2 mL of t h i s m i x t u r e . Four weeks l a t e r

t h e y were r e i m m u n i z e d w i t h t he same m i x t u r e p r e p a r e d w i t h i n c o m p l e t e F r u e n d s a d j u v a n t . The b o o s t e r i m m u n i -

z a t i o n s were r e p e a t e d e v e r y two w e e k s . T e s t b l e e d i n g s

were t a k e n one week a f t e r t he b o o s t e r s . A n t i s e r u m

w i t h s u i t a b l e t i t e r s w e r e o b t a i n e d by f o u r m o n t h s a f t e r

t h e i n i t i a l i m m u n i z a t i o n .

C. P r e p a r a t i o n of l a b e l e d c o n j u g a t e

N - [ 4 - ( 4 - A m i n o b e n z a m i d o ) - 4 - ( 2 - d i e t h y l a m i n o e t h y l ) b u t y l ] - 7 - S - g a l a c t o s y l c o u m a r i n - 3 - c a r b o x a m i d e .

To a s t i r r e d s u s p e n s i o n of 1 .4 g ( 3 . 8 mmol) o f

7 - P - g a l a c t o s y l c o u m a r i n - 3 - c a r b o x y l i c a c i d [ J . F . B u r d , . e t a l , CZin. Chem. 2 3 : 1 4 0 2 ( 1 9 7 7 ) ] in 40 mL of d r y

DMF was added 400 mg (4 mmol) t r i e t h y l a m i n e . The m i x -

t u r e was s t i r r e d u n t i l h o m o g e n e o u s , c o o l e d to - 1 0 ° C

u n d e r a r g o n , and c o m b i n e d w i t h 510 mg (4 mmol) of i s o -

b u t y l c h l o r o f o r m a t e . The r e a c t i o n was s t i r r e d at t h i s

t e m p e r a t u r e f o r 30 m i n u t e s to c o m p l e t e t h e f o r m a t i o n

of t he mixed a n h y d r i d e . A s o l u t i o n of t h e p r e c e e d i n g

d i a m i n e , ( 3 ) , Z = a m i n o , n = 4 [ 1 . 1 5 g, 3 .8 mmol] i n

25 mL of DMF was a d d e d and t h e r e a c t i o n a l l o w e d t o

s t a n d at 0°C f o r 12 h o u r s . C o n c e n t r a t i o n on a r o t a r y

e v a p o r a t o r a t t a c h e d to a vacuum pump l e f t a g l a s s y

To 1 .5 mL of t h e A n t i b o d y / E n z y m e R e a g e n t i n

c u v e t t e s was a d d e d 50 µL ( m i c r o l i t e r s ) of t he d i l u t e d

p r o c a i n a m i d e s t a n d a r d s . Then to b e g i n t h e r e a c t i o n ,

50 µL of t h e C o n j u g a t e R e a g e n t was a d d e d to e a c h

c u v e t t e w i t h m i x i n g . A f t e r 20 m i n u t e s t he f l u o r e s -

c e n c e i n t e n s i t y was m e a s u r e d in e a c h c u v e t t e ( e x c i t a -
