The CD40 antigen is a type I integral membrane protein of the tumor necrosis factorreceptor(TNFR)superfamily.ThisantigenisfoundonBcelllines,itisstronglyexpressedby interdigitatingcells (IDC),basalepithelialcells,and isalsopresentonmacrophages,someendothelialcells,andfolliculardendriticcells.Itisapan-Bmarker,absentonlyfromterminallydifferentiatedplasmacells.CD40 is implicated in the process of B cell selection in the germinal centre. Studiesdemonstrated that CD40 monoclonal antibodies induce strong homotypic adhesions inrestingBcellsand,togetherwithinterleukin-4(IL-4)maintainthecellcycleofblastsoftheBlineage.TheyalsocanpromotetheswitchtoIgEsecretion.ActivatedBcellsviaCD40antigeninthepresenceofIL-10differentiateintoplasmacellsandsecretelargeamountsof immunoglobulins.CD40bindstoCD154(CD40ligand)onTcells, resulting inBcellgrowthanddifferentiation.
