INTRODUCTION The Spirulina platensis is an excellent source of

phycocyanin , The protein fraction may contain up to 20% of phycocyanin

phycobiliproteins

4

Phycocyanin has been used mainly as a food pigment and cosmetic, recently , it’s also has shown the anti-inflammatory and anti-cancer properties

This paper show the effects of temperature and biomass-solvent ratio on phycocyanin concentration and extract purity to determine the optimum conditions for phycocyanin extraction

INTRODUCTION

MATERIAL AND METHOD Spirulina platensis LEB 52 different solvents for Phycocyanin extraction : distilled water

10 mM sodium phosphate bu er(pH 7.0)ff

10 mM sodium acetate bu er (pH 5.0)ff

0.15 M NaCl

CaCl2 10 g /L

Extraction procedure

mixing

dry biomass with solvent in a rotary

shaker at 30 °C

After centrifugation,

take the supernatant

Optical Density

was measured at 615 and 652 nm

Formula

where PC is the phycocyanin concentration (mg/ ml) OD615 is the optical density at 615 nm OD652 is the optical density at 652 nm

where EP is the extract purity

RESULT OF EXTRACTION WITH DIFFERENT SOLVENTS

pH and ionic strength lead to affect phycocyanin extraction

near the isoelectric point ( pH5.11)

protein interaction

extraction amount

It was observed that extraction with water can be a good choice

8

RESULT OF VARIABLE EFFECT ON PC & EP

temperaturebiomass / solvent

9

RESULT OF VARIABLE ANALYSIS

The variable biomass-solvent ratio strongly influenced the phycocyanin concentration

As ratio phycocyanin concentration

it makes the volume of extractant

10

RESULT OF VARIABLE ANALYSIS

PC model

The analysis of variance (ANOVA) was used to evaluate the adequacy of the fitThe F-value indicate the precision for the PC modle

11

temperature purity because high temperature facilitated the extraction of other proteins

CONCLUSION Water was chosen as the extractant, because it produced

high phycocyanin concentration and its low cost

The condition established for phycocyanin extraction was a biomass-solvent ratio of 0.08 g/ml, for 4 hrs, 25 °C

Under these conditions it’s possible to reach a phycocyanin concentration of 3.68 mg/ml and purity ratio (A615/A280) of 0.46

14

I can follow the extraction procedure and adjust the variables to see what is the result of phycocyanin concentration
