Confocal Detection Principle

Confocal Laser Scanning Raman Microscope hasbecome a widely recognized research instrumentin recent years. Confocal microscopy offersseveral advantages over conventional wide-fieldoptical microscopy, including the ability to controldepth of field, elimination or reduction ofbackground information away from the focal planeand the capability to collect serial optical sectionsfrom thick samples. The image of the extendedsample is generated by scanning the focused laserbeam across a defined area.

The pinhole aperture rejects the residual scatteredrays originated from any out-of-focus points on a sample.

PharmaceuticsConfocal Raman spectroscopy allows chemical compounds and

molecular conformers in various drugs to be identified and their

distribution mapped with high spatial resolution.

GeologyConfocal Raman microscopy is an excellent technique for

characterization of minerals, detection of components distribution

and their phase transitions.

CosmetologyConfocal Raman microspectroscopy is a promising technique which

enables measuring the skin care products as well as their penetration

capability.

ForensicsApplication areas include identification of unknown substances,

different types of fibers, glasses, paints, explosive materials, inks,

narcotic and toxic substances, proof of authenticity of documents.

Material scienceConfocal Raman offers excellent spatial resolution for characterization

of materials (superconductor, polymers, coatings, composites, carbon

nanotubes, graphene, etc.).

Heritage and Art, GemologyRaman spectroscopy allows identfication of pigments and binders

used in paintings. The spectroscopic analysis of archaeological

samples (ceramics, glasses, etc.) provides information on their origin

and history. Raman technique allows rapid identfication of colored

stones, natural and synthetic diamonds.

Major features

The smaller amount of illuminated pixels on the CCD matrix leads to

the smaller dark counts and the higher spectral resolution.

High spatial resolution and sensitivity

The highest spectral and imaging resolution withspecially designed spectrometer

Specially designed imaging spectrometer incorporates many features

that make it ideal for confocal Raman measurements. The image of

pinhole is projected to a multichannel detector without any aberrations.

Spectral resolution of NAµRAMS-01 with an Echelle grating-1

Recognition of living and dead cells

Raman spectra of the cell nucleus have significantly lower peaks in dead

cells as compared to living cells. A decrease in intensity is observed in

almost the full range, especially at the peaks of 783 and 827 1/cm and at

the peaks of 1003 and 1337 1/cm, which corresponds to the breaking of the

internal phosphodiester bonds in the DNA molecules and a decrease in

the level of protein expression.

New Age Instruments & Materials Pvt. Ltd.

However, in the nuclei of dead cells, lines increase by 1578 1/cm and new

Raman peaks appear at 1607 or 1114 1/cm, which corresponds to Phe and

Try, respectively. These results indicate the degradation of DNA molecules

and the presence of new aromatic amino acids rich in proteins, which

usually occur during cell apoptosis
