J.T.BakerYour professional guide through Solid Phase Extraction
3
IntroductionAmong chromatography suppliers,J.T.Baker stands out with a uniquebreadth of separations capabilities. You may know us for our high purityreagents, but we have also been inthe Liquid Chromatography (LC)business since the 1970s. In fact, wepioneered the development of SolidPhase Extraction (SPE).
We’re manufacturers of chromato-graphic media, developers of proprietary bonding chemistry, andproviders of products and know-howfor diverse separations techniques, scale and applications. Our productsfor analytical separations are focusedin three distinct applications:
• Analytical HPLC - WithBAKERBOND™ media for largemolecule or small molecule separations available in columnsor as bulk media.
• Flash Media and Cartridges - Our new, high-performancespherical silica media can befunctionalized with a variety ofsurface chemistries for optimalapplication performance.
• Solid Phase Extraction(both BAKERBONDconventional columnsand Speedisk™ high performance columns)- Our new 96-Wellplate, optimal for auto-
mated liquid handling systems, isthe most recent addition to theSpeedisk family. Our Speediskcolumns perform 9 times fasterthan conventional SPE columns.
Solid Phase Extraction for Sample PreparationWe are never satisfied with the status quo. Our improved BAKER-BOND spe products provide standard, reliable performance for today’sresearch, as well as QC pharma-ceutical, clinical and environmentalapplications. Our extraction tubedesign is optimized for SPE, andcompatible with manual and auto-mated SPE column processors. Ourproprietary column component formulations are ultraclean.
Furthermore, a wide range of BAKERBONDsorbents let you rationally choose thefunctionality that’sappropriate for yoursample.
The purpose of sample preparation
Sample clean up Necessary to eliminate impuritiesand/or isolate the component ofinterest from the matrix. This helpsto increase lifetime of the analyticalcolumn, prevents contamination ofequipment, thereby protectingexpensive instrumentation.
Sample concentration in order to reach the detection limits of the analytical equipment.Sample preparation selectively concentrates the components ofinterest prior to analysis.
Product characteristics
BAKERBOND spe Columns
With BAKERBOND spe columns, you can choose the solid phaseextraction column that best fitsyour sample size and performancerequirements. Use standardBAKERBOND spe 1, 3 and 6 mlultraclean polypropylene and glasscolumns, packed with 50-3000 mgsorbent, for samples from 0.2 to 10 ml
when standard speed, recovery, andfinal concentration with good economy are needed. • Optimized, round-rimmed tube
and frits made from ultrapurepolymers.
• Tubes fit into all popular processors and racks.
• Multi phase columns for specialenvironmental applications (e.g.PAH’s, PCB’s and Pesticides)
• Mixed Mode narc™ columns fordrugs of abuse sample preparation
• Wide Pore (300 Å) sorbents forbiological applications.
BAKERBOND Speedisk columns
J.T.Baker Speedisk columns reduceSPE run times by 60-80% and mayeliminate other time consumingsample preparation steps, such aspre-filtration and evaporation, some-times required with conventional SPEcolumns.
Thanks to the unique patentedSpeedisk laminar design andBAKERBOND high performancemicro-particles you not only getshorter run times, but also higher
capacity, optimum recovery andmore effective separation. The designis especially useful in clinical, biologicaland pharmaceutical drug applications.Speedisk columns are disposable, ultraclean, polypropylene columns(1-6 ml), pre-packed with high performance micro particles of 10µm(10-200 mg) and available in a widerange of sorbents (20 silica basedand 6 polymer based) to meet allyour separation needs.
‘Method Development Kits’ areavailable for BAKERBOND spe andSpeedisk columns. Please contact yourlocal office for more information.
Speedisk 96 Columns and Speedisk 96-well plates
BAKER spe- 12G Column Processor
Speedisk 96 columns multiply theadvantages of Speedisk columns by 96 to raise high throughput SPEto a higher level of performance. We offer these Speedisk products intwo different geometries. For methoddevelopment we offer the versatilemodular Speedisk 96 columns. Weinsert 96 “rimless” 20 mg Speediskcolumns in the Speedisk 96 columnholder and the assembly is ready forplacement on the Speedisk 96Positive Pressure Processor.

Fast, innovative & reliableSample preparation with J.T.Baker SPE technology
You can remove unneeded columnsfrom the holder, and assemble customtrays for efficient method develop-ment. Once method development iscomplete, use the Speedisk 96-wellplate, a one-piece, molded plate thatis pre-assembled with the sorbent ofyour choice. The new Speedisk 96-wellplate was designed with standardgeometries to adapt to popular auto-mated liquid handling systems.
Choose from more than 20 functiona-lities of silica and polymer-based sorbents for either Speedisk 96 columnsor the new Speedisk 96-well plates.
Positive Pressure Processors andAccessories for Extraction Columns
Speedisk 48 and 96 Positive Pressure Processor and
Speedisk 96 Column Holder
Speedisk 96 Positive Pressure Processor:The Speedisk Positive PressureProcessor enables you to combinethe advantages of a positive displace-ment processor with Speedisk columns. The processor produces the sameflow rate in each of the 96 columnsarrayed in 96 well plate.
Speedisk 48 Positive Pressure Processor:The Speedisk Positive PressureProcessor is perfect for processing 1,3 and 6 ml columns in batches of 1-48 samples. It distributes N2 orcompressed air to displace liquidfrom BAKERBOND spe columns or BAKERBOND Speedisk columnsand it easily enables a high controland precision.
Accessories for Extraction Columns
