EPIDERMISThis is the outer protective, non vascular layer of the skin.

DERMISThe dermis consists mostly of connective tissue and is much thicker than the epidermis.

SUBCUTANEOUS LAYER

The subcutaneous layer below the dermis consists of loose connective tissue and much fat. It acts as a protective cushion and helps to insulate the body by monitoring heat gain and heat loss.

Microsponge• The Microsponge Delivery System (MDS) is a unique technology for the controlled

release of topical agents

• Consists of macroporous beads, typically 10-25 microns in diameter, loaded with active agent

• When applied to the skin, the MDS releases its active ingredient on a time mode and also in response to other stimuli (rubbing, temperature, pH, etc)

• MDS technology is being used currently in cosmetics, over-the-counter (OTC) skin care, sunscreens and prescription products

• By delivering the active gradually to the skin, MDS-retinol, for example, has excellent efficacy with minimal irritation

Clinical Study: Journal of Dermatological Treatment (2000) LH Kligman and EH Gans

Once abandoned because of its instability, ROL can now be sequestered in Microsponge Polymers with low irritancy and cosmetic acceptability for treating the visible signs of aging and photoaging. Protected from oxidation by antioxidants such as vitamins C and E, it remains stable with a shelf life of at least 2 years. By obviating the rapid degradation that ROL would otherwise undergo, the new formulation has permitted the resumption of research with this agent. It was found that ROL was metabolized primarily to retinyl esters and to relatively small amounts of RA which activated nuclear RA receptors. ROL can be effective in inducing beneficial epidermal changes with less irritation than RA. **1.6% ROL induced significant epidermal thickening similar to that of 0.025% RA. Trace erythema, occurred with ROL whereas RA induced measurable erythema. ROL penetration was more effective than that of RA, but required a 10-fold higher concentration than RA. ROL increased epidermal thickness without the erythema induced by RA. Studies demonstrated that ROL has lower potency than RA, requiring higher concentrations than RA to produce similar epidermal effects. ROL is more effectively delivered into skin, inducing retinoid changes in the epidermis without irritation.

Liposome

• Liposomes are minute spherical oil droplets separated from an aqueous medium by a phospholipid bilayer

• The small size of liposomes enables them to pass through epidermis and act as a carrier for liquid substances, which are enclosed within the liposome’s hollow internal cavity

EnzymesAn enzyme is a protein molecule that is a biological catalyst with three characteristics:

1.The basic function of an enzyme is to increase the rate of a reaction. Most cellular reactions occur about a million times faster than they would in the absence of an enzyme.

2.Most enzymes act specifically with only one reactant (called a substrate) to produce products.

3.Enzymes are regulated from a state of low activity to high activity and vice versa.

Peptides• Peptide is a molecule formed by joining two or more amino acids

• They have the same peptide bonds as those in proteins, but are shorter in length • Peptides are named based on the number of amino acids in the chain: dipeptides (2),

tripeptides (3), tetrapeptides (4), pentapeptides (5) • When the number of amino acids is less than 50 these molecules are named peptides

while larger sequences are referred to as proteins • A peptide bond is a special linkage in which the nitrogen atom of one amino acid binds

to the carboxyl carbon atom of another

Lipopeptides• Lipopeptide is a peptide attached to a lipid (fat)

Antioxidants• An antioxidant is a molecule capable of inhibiting the oxidation of other molecules

• Oxidation is a chemical reaction that transfers electrons or hydrogen from a substance to an oxidizing agent

• Oxidation reactions can produce free radicals, these free radicals start chain reactions

• When the chain reaction occurs in a cell, it can cause damage or death to the cell

• Antioxidants terminate these chain reactions by removing free radical intermediates, and inhibit other oxidation reactions

• Vitamin A is a key vitamin that the skin needs topically.

• Oxidizes easily and is unstable.

• Oral consumption does not address the topical deficiency.

Retinol Cascade

• The retinol cascade is the process where special enzymes convert retinol to retinoic acid
