In this short course, we will review the basic concepts of ionization methods from both the structural and quantitative point of view, with an emphasis on mutual requirements between the ionization, the separation methods and the mass spectrometers design. The associated activation techniques leading to fragmentation
will be conceptually presented. Finally, the recently commercially available ion mobility separation and mass spectrometry imaging will be presented in terms of structural and analytical applications.

Strategies for efficient hyphenations of (U)HPLC with MS

Given the strongly different environments in which (U)HPLC and Mass Spectrometry take place (liquid phase, atmospheric pressure and above versus vacuum) they are in principle alien, incompatible methods.
Nevertheless early on in (U)HPLC system development, the method was successfully coupled online with MS. Since electrospray ionization has become commercially available, LC-MS evolved into a routine methodology.
However, MS detection puts constraints on any (U)HPLC system, the solvents used, the solute properties and sample preparation.
